Cover Story: Curcumin and its nanoformulations present a compelling multi-targeted strategy for glioblastoma therapy, addressing the limitations of conventional treatments. By modulating key oncogenic pathways such as PI3K/AKT/mTOR, NF-κB, and STAT3, curcumin enhances apoptosis, inhibits angiogenesis, and sensitizes tumor cells to temozolomide and radiation. Nanocarriers like liposomes, polymeric nanoparticles, and dendrimers enhance its solubility, bioavailability, and brain-targeted delivery. Preclinical studies support its efficacy alone and in combination with immunotherapy. Despite formulation and clinical translation challenges, nanocurcumin holds strong promise as a therapeutic adjunct in glioblastoma, with the potential to improve survival and quality of life in this devastating disease.
