Cover Story: The present review summarizes the multiple roles that the cellular prion protein (PrPC) plays in different cancer settings and highlights its role in the development and progression of epithelial cancers, including gastric, melanoma, breast, colorectal and pancreatic, as well as rare cancers. The review also suggests the consideration of the PrPC protein and its associated proteins as potential targets to treat resistance-associated cancers to conventional treatments. In addition, the review reports the molecular mechanisms by which PrPC triggers cancer development and/or contributes to its aggressiveness. The review strongly suggests that further comprehensive studies, focused on the discovery of PrPC-based therapy, may open up exciting new avenues for cancer research. View this paper
