Cover Story: The glycosylation of lipids results in the formation of glycosphingolipids (GSLs), which have been linked to cardiovascular disease. Diabetes leads to elevated GSL levels due to hyperglycemia and inflammation in adipose tissues. In this study, we isolated adiposomes (extracellular vesicles derived from adipose tissues) from obese diabetic patients and lean, healthy controls. We determined the GSL content of these adiposomes and their effects on the structure and function of endothelial cells. These adiposomes fused with endothelial cells in caveola-rich regions, resulting in a loss of integrity. We measured the upstream signaling pathways that lead to this effect and the downstream signaling pathways that become impacted by the loss of endothelial surface caveolae, such as nitric oxide production, a shear stress response, and albumin uptake.
