Cover Story: Of all the human copper-containing amine oxidases, human vascular adhesion protein-1 is a specific target in drug design for the treatment of inflammatory diseases and cancer, though off-targeting of diamine oxidase should be avoided. As part of the Special Issue “Design and Application of Metal-Binding Proteins” with Guest Editor Dr. Aimin Liu, we review the structural bioinformatics and structural biology approaches that can be employed to minimize the species-specific binding properties of vascular adhesion protein-1-targeted inhibitors and improve drug safety by avoiding their binding to diamine oxidase.
