Cover Story: Activation of NLRP3 is a complex process which is generally provoked by destabilization of cell homeostasis. Due to their shape, multiwalled carbon nanotubes (scattered in the cytoplasm of THP-1 macrophages) disrupted lysosomes. Released Cathepsin B subsequently activated NLRP3 assembly. On the other hand, pristine graphene platelets (mostly enclosed in endosomes) seemed “harmless”. However, their presence boosted stimulus from the present muramyl dipeptide, a representative of DAMPs. Such a synergic action led to strong NLRP3 activation. In this study, we have demonstrated unequivocal proinflammatory potential of carbon-based nanomaterials, which might be enhanced by various PAMPs/DAMPs. View this paper.
