Cover Story: Gut ischemia/reperfusion (I/R) injury, a severe and often fatal condition, releases damage-associated molecular patterns (DAMPs), such as extracellular cold-inducible RNA-binding protein (eCIRP). Milk fat globule–epidermal growth factor factor-VIII (MFG-E8)-derived oligopeptide 3 (MOP3) is a novel peptide enabling macrophage uptake of eCIRP via αvβ3-integrin. In our murine gut I/R model, the M2 macrophage population in mice decreased, while the M1 population increased without MOP3 treatment. However, with MOP3, the proportion of M2 macrophages rose while M1 declined. Through in vitro experimentation, M2 macrophage activity was driven by MOP3’s interaction with αvβ3-integrin, which mediates phagocytosis of eCIRP. This process, analogous to MFG-E8-induced efferocytosis, is a novel mechanism of MOP3. View this paper
