Cover Story: Transforming growth factor-β1 (TGF-β1) is a driver of wound healing and fibrosis that upregulates the profibrotic sialidase NEU3 by activating the RNA-binding protein DDX3 to increase NEU3 translation. NEU3 is an extracellular enzyme that releases active TGF-b1 from sequestration by latency-associated peptide (LAP). Here, we show that NEU3 levels in human lung fibroblasts increase within minutes of TGF-β1 exposure, that LAP alone also upregulates NEU3, and that LAP acts synergistically with TGF-b1. As a result, LAP + TGF-β1 upregulates NEU3 which then releases LAP + TGF-β1 from sequestration, and the positive feedback loop is blocked by NEU3 inhibitors and a DDX3 inhibitor. These findings reveal an unexpected signaling role for LAP and identify a rapid wound-healing response that, when dysregulated, may contribute to fibrosis. View this paper
