Cover Story: Tauopathies are marked by mitochondrial dysfunction, increasing neuronal vulnerability to degeneration. We investigated intercellular mitochondrial transfer (IMT) from astrocytes to neurons using SH-SY5Y/A172 co-cultures and human iPSC-derived cells. IMT was significantly enhanced in cells expressing P301L mutant tau, occurring mainly through contact-dependent mechanisms, correlating with increased tunneling nanotubes between cells. Tau pathology did not change the fate of transferred mitochondria once inside recipient cells. Importantly, these mitochondria remained functional, improving cellular respiration and bioenergetics in pathological cells. Altogether, IMT may represent a compensatory metabolic adaptation in tauopathy, sustaining neuronal bioenergetics via enhanced mitochondrial functions. View this paper
