Cover Story: Extensive research is being conducted on the therapeutic application of antimicrobial peptides due to the rise in antibiotic-resistant bacteria. Decisive criteria for the potential use of AMPs are antimicrobial efficacy, cytotoxicity, and biostability. While modifications to peptides can improve these properties, it is crucial to examine the impact of various modifications on each unique AMP. Recently, two new peptides, HT-16 and HT47, derived from the C-type lectin CLEC3A, have been found to be active against P. aeruginosa and S. aureus, but not MRSA. Mild cytotoxicity towards eukaryotic cells was observed for HT-47. Here, we modify HT-16 and HT-47 through truncation, the addition and substitution of amino acids, and the use of D‑amino acids to improve their antimicrobial properties as well as reduce their cytotoxicity. View this paper
