Cover Story: Malaria parasites reside within red blood cells and create multiple nested metabolic environments for heme production, utilization, and detoxification. In this review, we introduce the concept of “Russian dolls of heme metabolism,” in which three interconnected layers coexist within the infected erythrocyte: the residual heme biosynthetic capacity of the host red blood cell, the parasite food vacuole where hemoglobin is degraded and host heme is released, and the parasite’s own complete heme biosynthetic machinery. These compartments generate chemically distinct pools of heme and porphyrin intermediates that shape redox balance, artemisinin activation, and drug resistance. By viewing malaria through this compartmentalized framework, new therapeutic opportunities emerge that exploit vulnerabilities across nested heme metabolic networks. View this paper
