Cover Story: Climate change exposes plants to intensifying drought, salinity, and other abiotic stresses. Proline accumulation is widely used as a biochemical indicator of stress and is often interpreted as evidence of enhanced tolerance. However, this relationship is not universal. Depending on the species, genotype, stress intensity, and absolute concentration reached, proline may act as a functional osmolyte, ROS scavenger, molecular stabilizer, or signaling molecule—or may simply reflect physiological strain. This review challenges the proline-centric paradigm and argues that proline should be evaluated alongside other osmolytes and stress markers, using case-by-case and physiologically meaningful assessments. View this paper
