Cover Story: N4-(7-chloroquinolin-4-yl)-N1,N1-diethylpentane 1,4-diaminehydrochloride monohydrate (CQHCl·H2O) is the monohydrate chloride salt of the well-known antimalarial drug chloroquine, and it provides the first single-crystal structural evidence of a monoprotonated chloroquine species. The resolved crystal structure reveals a highly organized supramolecular architecture sustained by an extensive hydrogen-bonding network that links protonated chloroquine cations (CQH+), chloride anions, and lattice water molecules. Additionally, the work demonstrates the successful isolation of a pseudo-polymorphic form of a commercially available chloroquine salt through a heterogeneous crystallization approach, underscoring new possibilities in solid-form engineering of established pharmaceutical compounds.
