Cover Story: Colorectal cancer cells can evade apoptosis through anti-apoptotic BCL-2 family signaling and metabolic adaptation to cellular stress. This study identifies dimethyl fumarate (DMF) as a sensitizer of HCT-116 colorectal cancer cells to the BCL-2 inhibitor venetoclax. DMF disrupts mitochondrial respiration, causing membrane hyperpolarization, ATP depletion, and an autophagy-associated stress response. Combined with venetoclax, these effects are accompanied by reduced proliferation, sustained G2 cell-cycle arrest, and altered expression of cyclins and cyclin-dependent kinases (CDKs), culminating in enhanced cell death. By linking mitochondrial stress with impaired apoptotic resistance, the study identifies a clinically relevant repurposing strategy for further preclinical evaluation in colorectal cancer. View this paper
