Cover Story: Conserving poultry genetic resources requires reliable quality control tools to ensure that cryopreserved germplasm remains suitable for future breeding and restoration. We established baseline post‑thaw semen quality metrics for two conserved Norwegian rooster lines using computer‑assisted sperm analysis (CASA) and flow cytometric membrane integrity assessment. Frozen–thawed semen from 24 roosters was evaluated immediately after thawing and after short‑term refrigerated holding, revealing line‑specific differences, substantial individual variation, and reduced motility traits during holding. While objective assessments provide essential benchmarks for gene bank management, the disconnect between post‑thaw sperm quality and previously reported poor fertility highlights the need for complementary fertility studies and molecular biomarkers. View this paper
