Cover Story: Elevated potassium levels in the tumor microenvironment arise from necrotic cell death, during which the high intracellular potassium concentrations are released into the local environment. The resultant hyperkalemic environment has a particularly strong effect on immune cells—disrupting both T cell differentiation and tumor-associated macrophage polarization. Using an optical electrode as an exogenous contrast agent, our photoacoustic chemical imaging technology was able to generate functional images of the tumor microenvironment that map the potassium concentrations within. Inductively coupled plasma mass spectrometry of tumor interstitial fluid was used to verify the image’s potassium maps. We demonstrated a relationship between tumor size and potassium content, potentially reflecting the maturation of a necrotic tumor core. View this paper
