Cover Story: Perfusion cultivation coupled with kinetic modeling offers a promising strategy for intensifying microbial biomanufacturing while reducing media wastage. Here, we combined perfusion cultivation of Lactococcus lactis with kinetic modeling and Pareto-front analysis to balance recombinant FGF2 yield and media wastage. Model-guided perfusion increased biomass by 7.9-fold and FGF2 titers by 5-fold compared with batch cultivation. Reduced media wastage alongside enhanced product formation could lower operating costs and environmental impact. Higher biomass is attractive for probiotic manufacturing, while improved FGF2 yields enhance production economics. Together, these results highlight model-guided perfusion as a sustainable, cost-effective platform for microbial biomanufacturing. View this paper
