Cover Story: Disruption of cell adhesion is implicated in the pathology of the fungal metabolite, sporidesmin, which causes necrotizing inflammation of the biliary tract and liver in livestock that ingest the toxin. The toxicity of sporidesmin is associated with the reactivity of the sulfur atoms in a disulfide-bridged dioxopiperazine ring. Oxidative stress mechanisms based on cycles of reduction and oxidation of the sulfurs with a formation of reactive oxygen species is a commonly accepted toxic mechanism. In the current work, HepG2 cells cultured with sporidesmin showed a loss of adhesion with detachment of sheets of cells and individual cells in the absence of cell death or oxidative stress. We propose that sporidesmin interacts with proteins that regulate interactions between cells and with extracellular matrix, resulting in altered cell contact and subsequent tissue damage in vivo.
