Cover Story: The genus Gymnopilus has approximately 250 species described in the literature and biosynthesizes bioactive compounds, including polysaccharides. In the present study, polysaccharides composed mainly by β-glucans, produced by the mushroom Gymnopilus imperialis (from Atlantic Forest in Brazil), were evaluated in terms of in vitro immunomodulatory potential, iNOS expression levels, and NO production. Moreover, the gene expression profile of inflammatory modulators such as IL10, IL6, and TNF-α was also investigated to verify the macrophage phenotype induced by treatment. Interestingly, the nitric oxide production was significantly increased in treated samples, and the expression of inducible nitric oxide synthase (iNOS) showed that the fraction with β-glucans from G. imperialis induces M1 polarization phenotype.
