Special Issue Information

Dear Colleagues,

Mitochondria are at the core of multiple pathways within the cell. These organelles are fundamental in the coordination of cellular metabolism, stress response, apoptosis as well as immunity. Besides their central role in aerobic energy production, mitochondria are directly involved in several signal transduction pathways, which altogether act to determine cell fate and homeostasis. Alterations in mitochondrial biogenesis and metabolism contribute to the onset and progression of several pathologies. The gastrointestinal system occupies a central position in our body, regulating nutrients disposal, offering a first line of protection against pathogens and harbouring a microbial community, which has gained a lot of attention in the last years. Mitochondrial functions are essential to fulfil the great variety of aspects necessary to maintain an healthy gastrointestinal system. Perturbations of mitochondrial biogenesis and metabolism have been associated with a multitude of gastrointestinal disorders, including cancer, metabolic and inflammatory diseases. This Special Issue of IJMS will cover peculiar aspects of mitochondrial biogenesis and metabolism in relation to the gastrointestinal pathophysiology.

Inflammatory bowel disease (IBD) is one of the leading gut chronic inflammation disorders, especially prevalent in Western countries. Recent research suggests that mitochondria play a crucial role in IBD development and progression to the more severe disease—colorectal cancer (CRC). In this review, we focus on the role of mitochondrial mutations and dysfunctions in IBD and CRC. In addition, main mitochondria-related molecular pathways involved in IBD to CRC transition are discussed. Additionally, recent publications dedicated to mitochondria-targeted therapeutic approaches to cure IBD and prevent CRC progression are discussed. Full article
