Advances in Gene and Nucleic Acid-Based Therapeutics: From Molecular Innovations to Potential Misuse
This special issue belongs to the section "Molecular Genetics and Genomics".
Special Issue Information
Dear Colleagues,
Recent advances in gene and nucleic acid-based therapeutics have rapidly expanded the scope of modern biomedical research and clinical practice. Breakthroughs in mRNA lipid nanoparticle (LNP) technologies, genome editing platforms such as CRISPR/Cas systems, and diverse nucleic acid modalities—including antisense oligonucleotides, siRNA, and miRNA—have enabled novel approaches to treating genetic, infectious, and complex diseases. In parallel, improvements in viral and non-viral delivery systems, as well as cell-based therapies such as engineered immune cells, have accelerated translation from bench to bedside. As these technologies mature, their potential applicability is extending beyond traditional therapeutic boundaries, raising important scientific and regulatory questions regarding appropriate use, including distinctions between treatment and performance enhancement, such as gene doping.
This Special Issue aims to provide a comprehensive and balanced overview of the recent progress in gene and nucleic acid-based therapeutics, spanning fundamental, preclinical, and clinical research in both human and animal systems. We welcome contributions addressing molecular design, delivery technologies, pharmacokinetics, safety, and therapeutic applications, including mRNA-based vaccines, genome editing, and regenerative medicine. In addition, this Special Issue encourages studies on analytical and detection methodologies relevant to gene doping and gene transfer, such as PCR- and sequencing-based detection of exogenous genetic elements, biomarker discovery, and data-driven approaches for identifying non-natural genomic alterations. Perspectives on regulatory science and frameworks that support the appropriate and responsible application of these technologies are also within scope.

Manuscript Submission Information
Manuscripts should be submitted online at www.mdpi.com by registering and logging in to this website. Once you are registered, click here to go to the submission form. Manuscripts can be submitted until the deadline. All submissions that pass pre-check are peer-reviewed. Accepted papers will be published continuously in the journal (as soon as accepted) and will be listed together on the special issue website. Research articles, review articles as well as short communications are invited. For planned papers, a title and short abstract (about 250 words) can be sent to the Editorial Office for assessment.
Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-anonymized peer-review process. A guide for authors and other relevant information for submission of manuscripts is available on the Instructions for Authors page. International Journal of Molecular Sciences is an international peer-reviewed open access semimonthly journal published by MDPI.
Please visit the Instructions for Authors page before submitting a manuscript. There is an Article Processing Charge (APC) for publication in this open access journal. For details about the APC please see here. Submitted papers should be well formatted and use good English. Authors may use MDPI's English editing service prior to publication or during author revisions.
