Cover Story: Type-I, -II and -III interferons (IFNs) are crucial for the first line of cell-intrinsic host defense. While type-I (IFNα, IFNβ) and type-III (IFNλs) induce transcription of interferon-stimulated genes (ISGs) via the JAK/STAT pathway, type-II IFN (IFNγ) is secreted by T helper type-1 cells and fuels the adaptive immune response to the pathogen. In addition, the airway epithelial barrier is important for the host cell intrinsic defense and orchestrates antiviral immune response by producing cytokines such as IL-33. Using a genome-wide gene expression biosensor chip sensing the gene expression in organotypic 3D air-liquid interface cultures, we were able to show that all types of IFNs induced similar ISGs. Type-I and type-III IFN stimulation positively correlated with the expression of cell–cell adhesion and intercellular signaling, and IFNγ promoted cell proliferation. View this paper
