Viruses publishes Special Issues to create collections of papers on specific topics, with the aim of building a community of authors and readers to discuss the latest research and develop new ideas and research directions. Special Issues are led by Guest Editors, who are experts on the topic and all Special Issue submissions follow MDPI's standard editorial process. The journal’s Editor-in-Chief and/or designated Editorial Board Member will oversee Guest Editor appointments and Special Issue proposals, checking their content for relevance and ensuring the suitability of the material for the journal. The largest recorded global outbreak began in May 2022; with over 162; 000 cases across 140 countries (WHO; September 2025). Herein; we have analyzed a skin lesion sample from a male patient (June 2022); molecularly diagnosed as MPXV. The virus was isolated; purified; and characterized ultrastructurally; phenotypically; and genomically. In this study; three viral clones were obtained following phenotypic evaluation of MPXV lysis plaque characteristics over a three-course infection in BSC-40 cells. Unlike the vaccinia virus Western Reserve (VACV-WR) strain; MPXV clones did not form comet-like structures; suggesting reduced EEV particle release; which is associated with viral dissemination. Whole-genome sequencing and assembly identified subtle differences among clones. Comparative genomic analyses; including synteny and SNP calling; revealed intra-clonal differences and divergence from clade I and II references; although the variety of mutations found did not reveal possible variations at the protein level. Altogether; these findings provide novel insights into the MPXV replication cycle in vitro and the genomic characteristics of a strain isolated in Brazil during the early mpox outbreak.
