Dear Colleagues,
Viral proteases are essential for many viruses, driving polyprotein processing, virion maturation, and evasion of host immunity. Their central role makes them prime antiviral targets, successfully exploited in HIV, HCV, enteroviruses, monkeypox, and SARS-CoV-2 therapies.
This Special Issue will showcase the latest advances in antiviral protease inhibitor research, from drug repurposing and high-throughput assays to structure-guided or AI-driven design, resistance mechanisms, and combination therapies. We welcome original research and reviews, especially those offering fresh insights into inhibitor design, assay development, the structural biology of target–inhibitor interactions, antiviral mechanisms, and drug resistance.

Accepted papers will be published continuously in the journal (as soon as accepted) and will be listed together on the special issue website. Research articles, review articles as well as short communications are invited. For planned papers, a title and short abstract (about 100 words) can be sent to the Editorial Office for announcement on this website.
Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process. Viruses is an international peer-reviewed open access monthly journal published by MDPI.
