Cover Story: Type 2 diabetes is a public health concern worldwide. Biguanides like metformin (Met) are used as first-line drugs for the treatment of type 2 diabetes. Metformin can interact with the γ subunit of AMP-activated protein kinase (AMPK), a feature associated with the inhibition of gluconeogenesis and lipogenesis, thereby allowing for insulin to lower hepatic glucose production more effectively. Although metformin shows good activity, it causes side effects in the management of type 2 diabetes. In this sense, one strategy to improve biological activity by reducing negative effects is the formation of metallodrugs. Thus, this study evaluated the predictive physicochemical, pharmacokinetic, and toxicological profiles of metal–metformin complexes, while their interactions with the key metabolic regulator AMPK were explored through in silico tools. View this paper
