Cover Story: The coagulation Factor FXIIIA2B2 complex lies at a crucial junction between bleeding and thrombosis in the hemostatic order of the blood coagulation pathway. The absence of a biophysical/all atom structure for this complex limits the ability of coagulation scientists from designing putative inhibitors/drugs against this complex for related pathological states. It also limits the understanding of structural–functional aspects for this complex. To fill this void, we present in our current study the first all atom model of this complex built using integrative hybrid approaches that combine techniques of mass spectrometry, molecular modeling/docking, and atomic force microscopy. Furthermore, based on the structure and thermodynamic patterns, we present insights into the assembly and the activation induced disassembly of this complex. View this paper.
