Exosomes in Tumor Therapy: Molecular Mechanisms and Therapeutic Applications

Dear Colleagues,
Extracellular vesicles, particularly exosomes, have emerged as critical mediators of intercellular communication in malignant diseases. Tumor-derived exosomes regulate cancer cell growth, immune evasion, angiogenesis, metastatic dissemination, and resistance to pharmacological intervention through the transfer of proteins, lipids, and nucleic acids. In parallel, engineered or cell-derived exosomes are being actively examined as therapeutic carriers due to their biocompatibility, intrinsic targeting capacity, and ability to protect bioactive cargo. Recent studies published within the last seven years demonstrate that exosome-based strategies enable the delivery of small molecules, nucleic acids, and proteins to tumor tissues with reduced off-target toxicity. In addition, modulation of exosome production, cargo selection, and uptake has been shown to interfere with tumor progression and immune suppression. These findings position exosomes as both actionable therapeutic targets and adaptable delivery vehicles in oncology. This Special Issue aims to collect high-quality original research and authoritative reviews addressing the molecular composition, biogenetic regulation, functional consequences, and translational applications of exosomes in tumor therapy. Topics include, but are not limited to, exosome-mediated drug delivery, immune modulation, biomarker discovery, resistance mechanisms, and therapeutic intervention strategies. By assembling current experimental evidence and mechanistic insight, this Special Issue seeks to advance molecular understanding and therapeutic development in exosome-based cancer treatment.
