Neurodegenerative disorders are a group of chronic, incurable conditions characterized by the loss of function of nerve cells of the central or peripheral nervous system, resulting in progressive cognitive dysfunction, visual decline, motor deterioration, and eventually death. Despite numerous advances in the investigation of different aspects of the pathobiology of neurodegenerative conditions, the complex mechanisms triggering these diseases and driving their progression have yet to be fully elucidated. Mounting evidence indicates that neuroinflammation, alterations in mitochondrial function, and disturbances in redox homeostasis play a significant role in the pathogenesis of these disorders. These enhanced oxidative stress mechanisms, typically associated with a poor antioxidant response, constitute an attractive research paradigm to investigate the elements contributing to the pro-oxidant–antioxidant imbalance, identify the dysregulation of specific metabolic pathways, and explore novel therapeutic strategies. Bioenergetic shifts, increased mitochondrial production of reactive oxygen species, impaired Nrf2 central modulation of the antioxidant response, and imbalanced intake of antioxidant nutrients and vitamins that serve as enzymatic cofactors in the synthesis of cysteine and glutathione are some of the targets under investigation for creating a vicious cycle accelerating neurodegeneration in the development of chronic oxidative stress.
This Special Issue will focus on the pathogenic mechanisms leading to oxidative stress in different neurodegenerative disorders, including Alzheimer’s, Parkinson’s, and Huntington’s diseases, as well as amyotrophic lateral sclerosis and age-related eye pathologies, among others. The Special Issue provides insight into the interplay between the multifaceted molecular pathways linking metabolic/bioenergetic alterations, dietary contributors, and molecular stressors, providing an opportunity to explore novel antioxidant therapeutic strategies as disease-modifying approaches. Manuscripts in the form of original research articles, reviews, perspectives, and communications are welcome.

Accepted papers will be published continuously in the journal (as soon as accepted) and will be listed together on the special issue website. Research articles, review articles as well as short communications are invited. For planned papers, a title and short abstract (about 250 words) can be sent to the Editorial Office for assessment.
Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-anonymized peer-review process.
