Cover Story: Orthoflaviviruses are transmitted by mosquitoes and ticks and include major human pathogens such as dengue, Zika, yellow fever, West Nile, and tick-borne encephalitis viruses. Although licensed vaccines exist for a limited number of orthoflaviviruses, their expanding geographic distribution, driven by climate change and urbanization, underscores the urgent need for vaccines with a high safety profile and providing broad and durable protection. This review outlines the strengths and limitations of current and emerging orthoflavivirus vaccine platforms and discusses how advances in antigen design, presentation, and delivery can inform the development of next-generation orthoflavivirus vaccines. View this paper
