Cover Story: Camptothecin (CPT) exhibits anticancer activity, yet it is very hydrophobic with poor stability. In this study, a dual pH/thermo-responsive block copolymer of poly(acrylic acid-b-N-isopropylacrylamide) (PAA-b-PNP) was synthesized and applied to encapsulate CPT by self-assembly to form nanoparticles (NPs) above its cloud point. Chitosan was further applied on the surface for improving biocompatibility. At pH 2.0, these NPs could protect the CPT; yet at pH 6.0, they were internalized by Caco-2 cells, followed by the intracellular release of CPT. They became highly swollen at pH 7.4. Among several cancer cell lines, the highest cytotoxicity was observed for H460 cells. View this paper
