Cover Story: The increase in drug resistance in bacteria is an urgent global health issue. Amphiphilic α-hydrazido acid derivatives are structurally simple mimics of antimicrobial peptides. The mechanism of membrane permeabilization at subtoxic concentrations is due to a rapid insertion, followed by an accumulation into the bilayer that increases membrane fluidity and permeability without major structural damages. When evaluated in combination with conventional antibiotics, they proved to be synergistic with tetracycline against sensitive Staphylococcus aureus and Escherichia coli, as well as with ciprofloxacin and colistin against resistant strains. In other cases, additive effects were observed, with a remarkable increase in the activity of conventional antibiotics for combinations with ciprofloxacin toward susceptible S. aureus and methicillin toward MRSA. View this paper
