The rapid advancement of biomolecular therapeutics—including peptides, proteins, nucleic acids, and gene-editing tools—has revolutionized modern medicine, yet their clinical translation remains hindered by challenges in their stability, targeting, and controlled release. Smart delivery systems offer transformative solutions by integrating stimuli-responsive materials, nanotechnology, and bioengineering to protect therapeutic cargo, enhance cellular uptake, and enable precise spatiotemporal control. Innovations such as pH-sensitive nanoparticles, ligand-functionalized carriers, and stimuli-triggered hydrogels are being explored to overcome biological barriers and improve therapeutic indexes. This special topic collection highlights emerging strategies in smart delivery platforms tailored for biomolecular drugs, with emphasis on translational potential, regulatory perspectives, and disease-specific applications including cancer, genetic disorders, and autoimmune diseases.
