Cover Story: Our research incorporates the DNA–gold affinity interaction into an electrochemical nucleic-acid-based direct adsorption platform for the voltammetric detection of HOTAIR long noncoding RNA. Our platform utilizes highly specific and sensitive nanomaterials coupled with screen-printed gold electrodes. The assay features three main steps: (i) magnetic bead capture of HOTAIR targets, (ii) magnetic purification and isolation of targets, and (iii) direct adsorption and detection via interfacial affinity interaction. We demonstrate detection as low as 1 pM for samples containing designated proportions of synthetic HOTAIR spiked in buffer as well as healthy plasma samples. Our method shows huge potential in diagnostics due to its disposable, simple, inexpensive, easy-to-store, and portable design. View this paper
