Unmet Needs and Emerging Challenges in Multiple Myeloma
Topic Information
Dear Colleagues,
We are pleased to invite submissions for this Topic entitled "Unmet Needs and Emerging Challenges in Multiple Myeloma".
Multiple myeloma (MM) ranks as the second most prevalent hematological malignancy worldwide. Although substantial progress has been achieved in therapeutic options, multiple myeloma remains incurable. Key obstacles toward curative therapy include heterogeneity observed across phase III clinical trials and the lack of real‑world datasets that can reflect its broad clinical and molecular diversity. These limitations hinder accurate prognostic assessment and restrict the development of optimized personalized treatment strategies. Widely adopted prognostic systems such as R‑ISS integrate clinical and molecular parameters for risk stratification. Nevertheless, these existing models are associated with well‑recognized knowledge gaps and technical constraints. Moreover, they fail to capture important non‑genomic adverse prognostic factors, including extramedullary disease and circulating tumor cells. To enable clinicians to deliver appropriate risk‑adapted therapy for individual patients, comprehensive integration of clinical, biological and molecular signatures is urgently required. Advanced tools such as artificial intelligence (AI) offer promising approaches to achieve this goal. This Topic aims to gather recent research advances concerning the molecular pathogenesis of MM. It focuses on improved risk‑stratification frameworks, the integration of multi‑dimensional biological data, and emerging targeted therapies, with the ultimate objective of advancing personalized treatment toward becoming standard clinical practice for patients living with multiple myeloma.
Potential topics of interest include, but are not limited to:
- Application of genomics and proteomics in deciphering MM pathogenesis and drug‑resistance mechanisms, together with the identification of novel therapeutic targets and biomarker‑driven treatment approaches;
- Roles of multiple myeloma‑associated microbiomes in discovering novel biomarkers and therapeutic candidates;
- Implementation of bioinformatics and artificial intelligence (including machine learning) for big‑data and multi‑omics integration, as well as the construction of predictive models to support clinical management of MM patients;
- Precision‑medicine‑oriented strategies for patients with high progression risk, covering early‑intervention schemes and personalized therapeutic modalities;
- Clinical value of survival‑ and progression‑risk prediction for patient management and clinical outcomes, including risk‑adapted therapeutic strategies and quality‑of‑life assessment.
We welcome both original research articles and review papers. All submitted manuscripts will go through the standard peer‑review process.
