Cover Story: This figure illustrates the Calci-Inflammatory Network—a novel conceptual framework that redefines milk fever not simply as a calcium deficiency, but as an adaptive physiological response to systemic inflammation. The schematic details how lipopolysaccharide (LPS) from mucosal sites—such as the rumen, mammary gland, uterus, or hoof—triggers an inflammatory cascade via Toll-like receptor 4 (TLR4), leading to cytokine release that suppresses calcium regulatory pathways. The resulting hypocalcemia is sensed by calcium-sensing receptors (CaSRs) on immune cells, which attenuate further cytokine release. This negative feedback loop suggests that moderate hypocalcemia may serve a protective, anti-inflammatory function, challenging traditional paradigms and offering a holistic view of calcium–immune interactions in dairy cows. View this paper
