Cover Story: This study demonstrates that chronic opioid intake upregulates a distinct profile of plasma TNF superfamily (SF) molecules that are associated with immune cell interactions between Dendritic cells and T and B cells, which are important for antibody responses. In a study of people with HIV (PWH), those with associated opioid use disorder (OUD) exhibited higher post-flu vaccine antibody responses than PWH without OUD. The plasma profile in PWH+OUD was dominant in co-stimulatory TNF SF molecules, including 4-1BB, OX-40, CD40, CD30, and 4-1BBL and positively correlated with Flu Ab titers. In PWH without OUD, Ig SF molecules, including PDL-2, CTLA-4, and Perforin were dominant, with PDL-2 showing a negative correlation with Flu Ab titers. The role of opioid receptors and SF signaling pathways may reveal novel therapeutic strategies for immune-deficient states. View this paper
