Cover Story: Membrane proteins and lipids, which are integral to the biological membrane, work hand in hand to ensure the smooth running of a myriad of physiological processes. Understanding and observing the molecular interplay between lipids and proteins is an experimental challenge. This review presents the recent biophysical approaches aimed at understanding how membrane proteins and lipids interact. The role of lipids in modulating oligomerization, conformational dynamics and ligand binding is summarized, and different techniques to observe such modulation are introduced along the way. A specific emphasis is put on single-molecule FRET, cryoelectron microscopy and native mass spectrometry techniques, given their important contribution to the study of lipid–protein interactions in the last five years.
