Cover Story: Clinical trials for the HDAC inhibitors vorinostat, panobinostat, romidepsin, and pracinostat have failed in castration-resistant prostate cancers. The emergence of androgen-receptor-negative prostate cancers, protumorigenic HDACs, upregulation of p21 and P-glycoproteins, and a downregulation of histone acetyl transferases may be unexplored pharmacodynamic reasons for this failure. Considerations of these key areas are likely to be vital in upcoming clinical trials. View this paper.
