Cover Story: Radioresistance is a major barrier to achieving efficient local tumor control. The combination of radiotherapy (RT) with immunotherapy has improved outcomes for patients with solid tumors. It becomes important to understand how radiation modulates immune mechanisms of radioresistance. We summarize how radiation modulates the expression and activation of Toll-Like Receptors in the tumor microenvironment. Radiation induces the release of damage associated molecular patterns from damaged or dying cells. DAMPs can be recognized by pattern recognition receptors such as TLRs. This triggers a signaling cascade that results in a pro- or anti-tumor effect, depending on the specific TLR and the type of cancer. The rational combination of TLR-based therapies with RT can enhance tumor recognition by antigen-presenting cells and lead to sustained antitumor immunity and effective local tumor control. View this paper
