Cover Story: C. albicans Prn1 is a protein that is similar to mammalian Pirin, which increases in response to H2O2, a compound used to mimic the oxidative action of phagocytes. To analyse the function of Prn1 in response to oxidative stress, the deleted mutant strain prn1∆ and the wild-type strain SN250 were treated with H2O2. Proteomics studies showed important differences in proteins with oxidoreductase activity, proteasome-related proteins, and several important transcription factors, such as Mnl1. Functional studies validated the results of proteomics studies and showed that the absence of Prn1 led to higher ROS levels, decreased survival, increased apoptosis, and increased proteasome activity. Results support the important role of Prn1 under oxidative stress and its possible role as a target for new antifungals. View this paper
