Cover Story: A pH-responsive theranostic agent based on core cross-linked (CCL) micelles was constructed via thiol–epoxy click chemistry between thiol-terminated tetraphenylethylene derivatives (TPE-4SH) and glycidyl moieties. The resulting CCL micelles were further functionalized: A targeting motif, pH low insertion peptide (pHLIP), and a magnetic resonance (MR) contrast agent were loaded with chemotherapeutic camptothecin (CPT). The as-prepared theranostic vector was efficiently taken up by taking advantage of the pHLIP-facilitated internalization that can be facilely traced by TPE fluorescence and MR imaging. Selective intracellular CPT release was accomplished, due to a hydrophobic-to-hydrophilic transition of micelle cores within acidic organelles. By Jinming Hu. View this paper.
