Cover Story: A viral protein R-derived 7-mer peptide with photo-reactive benzoylphenylalanyl and biotin residues that exhibits low micromolar HIV-1 integrase (IN) inhibitory potency, was shown to bind directly to IN during photo-crosslinking experiments. The peptide does not interfere with IN–DNA interactions or induce higher-order aberrant IN multimerization, suggesting a mode of action distinct from clinically used integrase strand transfer inhibitors (INSTIs) and experimental allosteric IN inhibitors. This pharmacological tool may aid identifying a site for therapeutic development. View this paper.
