A series of rivastigmine hybrids, incorporating rivastigmine fragments (RIV) and a set of different antioxidant scaffolds, were designed, synthesized, and evaluated as multifunctional agents for the potential therapy of Alzheimer’s disease (AD). In vitro bioactivity assays indicated that some compounds have very good antioxidant (radical-scavenging) activity. The compounds also displayed good inhibitory activity against cholinesterases, though the bigger-sized hybrids showed higher inhibitory ability for butyrylcholinesterase (BChE) than for acetylcholinesterase (AChE), due to the larger active site cavity of BChE. All the hybrids exhibited an inhibition capacity for self-induced amyloid-β (Aβ1–42) aggregation. Furthermore, cell assays demonstrated that some compounds showed capacity for rescuing neuroblastoma cells from toxicity induced by reactive oxygen species (ROS). Among these RIV hybrids, the best in vitro multifunctional capacity was found for the caffeic acid derivatives enclosing catechol moieties (4AY5, 4AY6), though the Trolox derivatives (4AY2, 4BY2) presented the best cell neuroprotective activity against oxidative damage. Molecular-docking studies provided structural insights into the binding modes of RIV-based hybrids to the cholinesterases, revealing key interaction patterns despite some lack of correlation with inhibitory potency. Overall, the balanced multifunctional profiles of these hybrids render them potentially promising candidates for treating AD, thus deserving further investigation. Full article

Viticis Fructus (VF), a fruit known for its unique flavor profile and various health benefits, demonstrates substantial quality variations depending on its area of production. Traditional methods of production area verification based on internal compound analysis are hampered by a number of technical limitations. This investigation systematically characterized the cuticular wax composition of VF sample from a diverse variety of production areas. Quantitative analyses were conducted to evaluate the spatial distribution patterns of the wax constituents. Significant regional variations were observed: Anhui sample exhibited the highest total wax content (21.39 μg/cm2), with n-alkanes dominating at 76.67%. High-latitude regions showed elevated triterpenoid acid levels, with maslinic acid (0.53 μg/cm2) and ursolic acid (0.34 μg/cm2) concentrations exceeding those of their low-latitude counterparts by four- and three-fold, respectively. Altitudinal influence manifested in long-chain alcohol accumulation, as triacontanol reached 0.87 μg/cm2in high-altitude sample. Five key biomarkers demonstrated direct quality correlations: eicosanoic acid, n-triacontane, dotriacontanol, β-amyrin, and α-amyrin. This study established three novel origin identification protocols: single-component quantification, multi-component wax profiling, and wax ratio analysis. This work not only reveals the latitudinal dependence of VF wax composition, but also provides a scientific framework for geographical authentication. Our findings advance wax-based quality evaluation methodologies for fruit products, offering practical solutions for production area verification challenges in food raw materials. Full article

The food industry consumes large amounts of water across various processes, and generates wastewater characterized by parameters like biochemical oxygen demand, chemical oxygen demand, pH, suspended solids, and nutrients. To meet environmental standards and enable reuse or valorization, treatment methods such as physicochemical, biological, and membrane-based processes are applied. This review focuses on the valorization of food industry wastewater in the biotechnological production of high-value products, with an emphasis on starch-rich wastewater, wineries and confectionery industry wastewater, and with a focus on new technologies for reduces environmental burden but also supports circular economy principles. Starch-rich wastewaters, particularly those generated by the potato processing industry, offer considerable potential for biotechnological valorization due to their high content of soluble starch, proteins, organic acids, minerals, and lipids. These effluents can be efficiently converted by various fungi (e.g., Aspergillus, Trichoderma) and yeasts (e.g., Rhodotorula, Candida) into value-added products such as lipids for biodiesel, organic acids, microbial proteins, carotenoids, and biofungicides. Similarly, winery wastewaters, characterized by elevated concentrations of sugars and polyphenols, have been successfully utilized as medium for microbial cultivation and product synthesis. Microorganisms belonging to the genera Aspergillus, Trichoderma, Chlorella, Klebsiella, and Xanthomonas have demonstrated the ability to transform these effluents into biofuels, microbial biomass, biopolymers, and proteins, contributing to sustainable bioprocess development. Additionally, wastewater from the confectionery industry, rich in sugars, proteins, and lipids, serves as a favorable fermentation medium for the production of xanthan gum, bioethanol, biopesticides, and bioplastics (e.g., PHA and PHB). Microorganisms of the genera Xanthomonas, Bacillus, Zymomonas, and Cupriavidus are commonly employed in these processes. Although there are still certain regulatory issues, research gaps, and the need for more detailed economic analysis and kinetics of such production, we can conclude that this type of biotechnological production on waste streams has great potential, contributing to environmental sustainability and advancing the principles of the circular economy. Pectin is a multifunctional polysaccharide whose structural attributes, including degree of esterification (DE), molecular weight (MW), and branching, directly affect its gelling, emulsifying, and bioactive properties. Conventional pectin extraction relies on acid- or alkali-based methods that degrade the pectin structure, generate chemical waste, and alter its physicochemical and functional properties. Although novel methods such as ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), and enzyme-assisted extraction (EAE) are recognized as environmentally friendly alternatives, they frequently use acids or alkalis as solvents. This review focuses on pectin extraction methods that do not involve acidic or alkaline solvents such as chelating agents, super/subcritical water, and deep eutectic solvents (DESs) composed of neutral components. This review also discusses how these alternative extraction methods can preserve or modify the key structural features of pectin, thereby influencing its monosaccharide composition, molecular conformation, and interactions with other biopolymers. Furthermore, the influence of these structural variations on the rheological properties, gelling behaviors, and potential applications of pectin in the food, pharmaceutical, and biomedical fields are discussed. This review provides insights into alternative strategies for obtaining structurally intact and functionally diverse pectin by examining the relationship between the extraction conditions and pectin functionality. The article presents the first method based on high-performance liquid chromatography and ultraviolet detection (HPLC-UV) for the determination of timonacic (thioproline, 1,3-thiazolidine-4-carboxylic acid, tPro) in pharmaceutical tablets and face care products (creams, sera, foundations, suncreams). Sample preparation primarily involves solid-liquid extraction (SLE) of tPro with 0.2 mol/L phosphate buffer pH 6, derivatization with 0.25 mol/L 2-chloro-1-methylquinolinium tetrafluoroborate (CMQT), followed by polytetrafluoroethylene (PTFE) membrane filtration. The chromatographic separation of the stable UV-absorbing 2-S-quinolinium derivative is achieved within 14 min at 25 °C on a Zorbax SB-C18 (150 × 4.6 mm, 5 µm) column using gradient elution. The eluent consists of 0.1 mol/L trichloroacetic acid (TCA), pH 1.7, in a mixture with acetonitrile (ACN) delivered at a flow rate of 1 mL/min. The analyte is quantified by monitoring at 348 nm. The assay linearity was observed within 0.5–125 μmol/L. The limit of quantification (LOQ) was found to be 0.5 μmol/L. The accuracy ranged from 93.22% to 104.31% and 97.38% to 103.48%, while precision varied from 0.30% to 11.23% and 1.13% to 9.64% for intra- and inter-assay measurements, respectively. The method was successfully applied to commercially available on the Polish market pharmaceutical and cosmetic products. Full article

In the search for alternative protein sources, single cell proteins have gained increasing attention in recent years. Among them, proteins derived from yeast represent a promising but still underexplored option. To enable their application in food product design, their techno-functional properties must be understood. In order to investigate the impact of precipitation pH on their emulsion-stabilizing properties, yeast proteins from Saccharomyces cerevisiae were isolated via precipitation at different pH (pH 3.5 to 5) after cell disruption in the high-pressure homogenizer. Emulsions containing 5 wt% oil and ~1 wt% protein were analyzed for stability based on their droplet size distribution. Proteins precipitated at pH 3.5 stabilized the smallest oil droplets and prevented partitioning of the emulsion, outperforming proteins precipitated at higher pH values. It is hypothesized that precipitation under acidic conditions induces protein denaturation and thereby exposes hydrophobic regions that enhance adsorption at the oil–water interface and the stabilization of the dispersed oil phase. To investigate the stabilization mechanism, the molecular weight of the proteins was determined using SDS-PAGE, their solubility using Bradford assay, and their aggregation behavior using static laser scattering. Proteins precipitated at pH 3.5 possessed larger molecular weights, lower solubility, and a strong tendency to aggregate. Overall, the findings highlight the potential of yeast-derived proteins as bio-surfactants and suggest that pH-controlled precipitation can tailor their functionality in food formulations. This research investigated the impact of corn–soybean meal-based fermented feed on the growth performance, pork quality, and fatty acid profiles of lean-type finishing pigs. A total of 80 lean-type growing DLY (Duroc × Landrace–Yorkshire) pigs were randomly assigned to 2 groups, with 5 replicates of 8 pigs per pen. The pigs in control group (CON group) were fed a basal diet, while the pigs in fermented feed group (FF group) were fed a diet supplemented with 10% fermented feed. The experimental period lasted 70 days. Results exhibited that pigs in FF group had a significant increase in final body weight and average daily gain (ADG) (p < 0.05) and had a significant decrease in the feed-to-gain ratio (F/G) (p < 0.05). The FF group also exhibited significant promotion in muscle intramuscular fat content, marbling score, and meat color and significantly reduced the meat shear force and drip loss (p < 0.05). Serum analysis indicated that fermented feed significantly elevated blood glucose, total cholesterol, triglyceride levels, and serum hormones such as insulin, leptin, and IGF-1 (p < 0.05). Additionally, fermented feed significantly elevated the levels of polyunsaturated fatty acids (PUFAs) and monounsaturated fatty acids (MUFAs), whereas it decreased the saturated fatty acids (SFAs) contents (p < 0.05). The fermented feed also significantly enhanced pork nutritional values (p < 0.05). The fermented feed increased the expression of IGF-1, SREBP1c, PDE3, PPARγ, SCD5, and FAT/CD36 mRNA (p < 0.05). Furthermore, microbial 16S rDNA analysis uncovered that FF supplementation significantly reduced the Campilobacterota phylum abundance, while increasing the genus abundances of Clostridium_sensu_stricto, norank_f_Oscillospiraceae, unclassified_c_Clostridia, and V9D2013 (p < 0.05). In summary, the results indicated that the microbial fermented feed exhibited the regulation effects on pork quality and nutritional values of lean-type pigs through regulating lipid metabolism and gut microbial composition. The increasing demand for sustainable pet-food solutions has driven interest in alternative protein sources, as researchers seek to avoid allergenic foods while maintaining optimal pet nutrition. This review explores recent scientific publications, patent trends, and market trends relating to various alternative protein sources, including plant-based, aquatic, insect-derived, and cell-based sources. Their nutritional composition, functional properties, and potential benefits for pet health were assessed. Plant-based proteins, such as soy, pea, and lentils, provide essential amino acids and functional properties suitable for meat analogues. Microalgae and seaweed offer rich sources of omega-3 fatty acids, antioxidants, and bioactive compounds. Insect-based proteins such as black-soldier-fly larvae and mealworms are highly digestible and rich in essential nutrients, with additional benefits for gut health. Emerging cell-based proteins present a novel, lab-grown alternative with promising sustainability and nutritional advantages. While these protein sources offer significant benefits, challenges related to digestibility, palatability, regulatory approval, and consumer acceptance must be addressed. The emphasis of the present research is on current developments for industry uses and future potential. The analysis sheds light on the contributions of alternative protein sources to the promotion of sustainable and nutrient meals for pets. Full article

Background: Spinal muscular atrophy type 1 (SMA1) is a severe neuromuscular disorder characterized by progressive muscle weakness and atrophy, including the muscles of the oral cavity and esophagus. Eosinophilic esophagitis (EoE), a chronic, allergic disease, presents with eosinophilic infiltration of the esophagus, leading to esophageal dysmotility. Feeding difficulties may occur in both conditions. So far, the coexistence of EoE and SMA1 has not been described; we present the first such case. Case presentation: The patient was a girl with SMA1 diagnosed shortly after birth, treated with nusinersen and onasemnogene abeparvovec, and fed a standard industrial diet through a gastrostomy. In her second year of life, she developed increasing symptoms: distress during feeding, regurgitation, vomiting, and weight loss. She was treated with proton pump inhibitors without clinical improvement. Gastroscopy was performed, revealing superficial epithelial damage with bleeding in the proximal esophagus. Histopathology showed chronic inflammation with up to 150 eosinophils per high-power field, microabscesses, spongiosis, and basal layer hypertrophy. The girl was diagnosed with EoE. Her diet was switched from a standard industrial formula to an amino acid-based formula, which led to marked clinical improvement, the resolution of symptoms, and appropriate weight gain. Conclusions: This case report highlights the challenges of diagnosing EoE in SMA1 patients and emphasizes the need for multidisciplinary approaches and further investigation of allergic manifestations in SMA1 patients. Activation of Peracetic Acid by Ozone for Recalcitrant Pollutant Degradation: Accelerated Kinetics, Byproduct Mitigation, and Microbial Inactivation

Iopamidol (IPM), as a typical recalcitrant emerging pollutant and precursor of iodinated disinfection by-products (I-DBPs), is unsuccessfully removed by conventional wastewater treatment processes. This study comprehensively evaluated the ozone/peracetic acid (O3/PAA) process for IPM degradation, focusing on degradation kinetics, environmental impacts, transformation products, ecotoxicity, disinfection byproducts (DBPs), and microbial inactivation. The O3/PAA system synergistically activates PAA via O3 to generate hydroxyl radicals (OH) and organic radicals (CH3COO and CH3CO(O)O), achieving an IPM degradation rate constant of 0.10 min−1, which was significantly higher than individual O3 or PAA treatments. The degradation efficiency of IPM in the O3/PAA system exhibited a positive correlation with solution pH, achieving a maximum degradation rate constant of 0.23 min−1 under alkaline conditions (pH 9.0). Furthermore, the process demonstrated strong resistance to interference from coexisting anions, maintaining robust IPM removal efficiency in the presence of common aqueous matrix constituents. Furthermore, quenching experiments revealed OH dominated IPM degradation in O3/PAA system, while the direct oxidation by O3 and R-O played secondary roles. Additionally, based on transformation products (TPs) identification and ECOSAR predictions, the primary degradation pathways were elucidated and the potential ecotoxicity of TPs was systematically assessed. DBPs analysis after chlorination revealed that the O3/PAA (2.5:3) system achieved the lowest total DBPs concentration (99.88 μg/L), representing a 71.5% reduction compared to PAA alone. Amongst, dichloroacetamide (DCAM) dominated the DBPs profile, comprising > 60% of total species. Furthermore, the O3/PAA process achieved rapid 5–6 log reductions of E. coli. and S. aureus within 3 min. These results highlight the dual advantages of O3/PAA in effective disinfection and byproduct control, supporting its application in sustainable wastewater treatment. Full article

Renewable fuels produced from animal- and plant-based edible oils have emerged as an alternative to oil and natural gas. Burgeoning interest in renewables can be attributed to the rapid depletion of fossil fuels caused by the global energy demand and the environmental advantages of renewables, specifically reduced emissions of greenhouse gases. An important property of the feedstock that is crucial for the conversion of edible oils to renewable fuels is the total acid number (TAN), as even a small increase in TAN for the feedstock can lead to corrosion of the catalyst in the refining process. Currently, the TAN is determined by potentiometric titration, which is time-consuming, expensive, and requires the preparation of reagents. As part of an effort to promote the use of renewable fuels, a partial least squares regression method with orthogonal signal correction to remove spectral information related to the sample background was developed to determine the TAN from the mid-infrared (IR) spectra of the feedstock. Digitally blended mid-IR spectral data were generated to fill in regions of the PLS calibration where there were very few samples. By combining experimental and digitally blended mid-IR spectral data to ensure adequate sample representation in all regions of the spectra–property calibration and better understand the spectra–property relationship through the identification of sample outliers in the original data that can be difficult to detect because of swamping, a PLS regression model for TAN (R2 = 0.992, cross-validated root mean square error = 0.468, and bias = 0.0036) has been developed from 118 experimental and digitally blended mid-IR spectra of commercial feedstock. Thus, feedstock whose TAN value is too high for refining can be flagged using the proposed mid-IR method, which is faster and easier to use than the current titrimetric method. Full article

Metabotype Risk Clustering Based on Metabolic Disease Biomarkers and Its Association with Metabolic Syndrome in Korean Adults: Findings from the 2016–2023 Korea National Health and Nutrition Examination Survey (KNHANES)

Background: Metabolic syndrome (MetS) is a multifactorial condition involving central obesity, dyslipidemia, hypertension, and impaired glucose metabolism, significantly increasing the risk of type 2 diabetes and cardiovascular disease. Objectives: Given the clinical heterogeneity of MetS, this study aimed to identify distinct metabolic phenotypes, referred to as metabotypes, using validated biomarkers and to examine their association with MetS. Materials and Methods: A total of 1245 Korean adults aged 19–79 years were selected from the 2016–2023 Korea National Health and Nutrition Examination Survey. Metabotype risk clusters were derived using k-means clustering based on five biomarkers: body mass index (BMI), uric acid, fasting blood glucose (FBG), high-density lipoprotein cholesterol (HDLc), and non-HDL cholesterol (non-HDLc). Multivariable logistic regression was used to assess associations with MetS. Results: Three distinct metabotype risk clusters (low, intermediate, and high risk) were identified. The high-risk cluster exhibited significantly worse metabolic profiles, including elevated BMI, FBG, HbA1c, triglyceride, and reduced HDLc. The prevalence of MetS increased progressively across metabotype risk clusters (OR: 5.46, 95% CI: 2.89–10.30, p < 0.001). In sex-stratified analyses, the high-risk cluster was strongly associated with MetS in both men (OR: 9.22, 95% CI: 3.49–24.36, p < 0.001) and women (OR: 3.70, 95% CI: 1.56–8.75, p = 0.003), with notable sex-specific differences in lipid profiles, particularly in HDLc. Conclusion: These findings support the utility of metabotyping using routine biomarkers as a tool for early identification of high-risk individuals and the development of personalized prevention strategies in clinical and public health settings. With a rapidly growing incidence and prevalence, Alzheimer’s disease (AD) is rapidly becoming one of the most disabling, lethal, and expensive diseases of the century. To diagnose AD as early as possible, the scientific world struggles to find reliable and non-invasive biomarkers that could predict the conversion of mild cognitive impairment to AD and delineate the ongoing pathogenic vicious pathways to be targeted with therapy. Research supports the use of blood biomarkers, such as Aβ1-42/Aβ1-40 ratio, phosphorylated tau181, and p-tau217 for diagnostic purposes, although the cut-offs are not clearly established and can depend on the assays used. For more accurate diagnosis, markers of neurodegeneration (neurofilament light) and neuroinflammation (glial fibrillary acidic protein) could be introduced in the biomarker panel. The recent approval of the Lumipulse G p-tau217/Aβ1-42 plasma ratio by the FDA for the early detection of amyloid plaques associated with Alzheimer’s disease in adult patients, aged 55 years and older, exhibiting signs and symptoms of the disease represents a significant advancement in the diagnosis of Alzheimer’s disease, offering a more accessible and less invasive way to diagnose this devastating disease and allow potentially earlier access to treatment options. Radical TTM-DMODPA for Ascorbic Acid Non-Catalytic Visual Detection

Ascorbic acid (AA) plays a multidimensional role in human physiological and pathological processes, and the detection of its urinary concentration facilitates the diagnosis of metabolic or kidney diseases. Visual detection exhibits minimal reliance on instrumentation and is suitable for on-site analysis in routine settings. Current visual colorimetric detection methods typically rely on enzymatic or nanozyme-based catalysis. Organic neutral radicals bearing unpaired electrons represent a class of materials exhibiting intrinsic responsiveness to redox stimuli. The tris (2,4,6-trichlorophenyl) methyl (TTM) radical has attracted widespread attention for its adjustable optical properties and sensitive response to external redox stimuli. We synthesized a novel radical TTM-DMODPA and applied it for non-catalytic colorimetric detection of AA. It not only enables quantitative AA measurement via UV-vis spectroscopy (linear range: 1.25–75 μmol/L, LOD: 0.288 μmol/L) but also facilitates instrument-free visual detection using smartphone cameras (linear range: 0–65 μmol/L, LOD: 1.46 μmol/L). This method demonstrated satisfactory performance in the measurement of AA in actual urine samples. Recovery rates ranged from 97.8% to 104.1%. Consequently, this work provides a portable and effective method for assessing AA levels in actual urine samples. Full article

Impact of Abiotic Stress-Reducing Cultivation Technologies and Long-Term Storage on the Oxidative Potential of Edible Potato Tubers (Solanum tuberosum L.)

Currently, in the context of the emphasis on introducing a reduction in mineral fertilization and the increase in pressure on sustainable agriculture, magnesium fertilization and the use of biostimulants are becoming an alternative tool to increase the quality of potato tuber yield. This study aimed to assess the impact of potato genotype, cultivation technology, and long-term storage on the susceptibility of tubers to enzymatic browning. Two edible potato varieties were examined: the early ‘Wega’ and the mid-early ‘Soraya’. It was demonstrated that the varieties maintained their characteristic browning susceptibility consistent with their breeding descriptions. The ‘Wega’ variety exhibited decreasing browning susceptibility immediately after harvest; however, after 6 months of storage, its susceptibility significantly increased, exceeding that of the ‘Soraya’ variety. Additionally, the application of magnesium fertilization (90 kg ha−1) and biostimulant treatment (3 L ha−1) most effectively reduced the oxidative potential of the tubers, thereby decreasing browning susceptibility. This is due to a significant change in the concentration of organic acids responsible for enzymatic browning processes. A decrease in the content of chlorogenic acid by 9.4% and 8.4% and an increase in the content of citric and ascorbic acid by 11.1%, 5.3%, and 13.6% were achieved. Storage significantly affected the chemical composition of the tubers. An increase in chlorogenic (7.3%) and citric (5.8%) acids and a decrease in ascorbic (34%) acid content were observed. These changes correlated with the intensification of browning, with the increase in chlorogenic acid and the decrease in ascorbic acid having the greatest influence. The results indicate that the technology based on supplementary fertilization and biostimulation improves the quality of potato raw material without a significant increase in production costs. Further research on varieties with different vegetation lengths and those intended for food processing and starch production is advised. Full article
