Cover Story: Besides insulin secretion stimulation, incretin-based therapies improve glycemic control due to inhibition of gastric acid secretion, delay gastric emptying, inhibit gut motility and induce satiety. We aimed to understand their role in modulating gastric fundus motility and the mechanisms involved in control and type 2 diabetic rats. Liraglutide inhibited the relaxation induced by SNP and NA, without affecting nNOS activity. In isolated rat gastric fundus pre-contracted with carbachol, GLP-1 receptor agonists liraglutide and exenatide induced receptor-dependent contractile responses. Thus, Liraglutide-induced increased gastric muscle tone may reduce gastric accommodation during food intake. This may contribute to the significant decrease in caloric intake and body weight in diabetic rats, being a peripheral mechanism involved in satiety. View this paper
