Cover Story: Treatment with immunotherapeutic modalities has shown long-term tumor regression in a minority of patients with glioblastoma—the most common and malignant form of primary brain cancer in adults. Chokshi et al. provide a comprehensive review of recent clinical and preclinical efforts to evaluate vaccination strategies, immune checkpoint inhibitors, and chimeric antigen receptor T cells. The authors describe the mechanisms of therapy evasion to these modalities and efforts to overcome resistance, with a goal to provide durable responses for the majority of glioblastoma patients.
