The strength mobilisation framework was adopted for the first time to describe the stress–strain responses for three different types of sands, including a total of 30 published drained triaxial tests—25 for Karlsruhe Fine Sand, 2 for Ottawa sands and 3 for Fontainebleau sand, under confining pressures ranging from 50 to 400 kPa. The peak shear strength τpeak obtained from drained triaxial shearing of these sands was used to normalise shear stress. Shear strains normalised at peak strength γpeak and at half peak of shear strength γM=2 were taken as the normalised reference strains, and the results were compared. Power–law functions were then derived when the mobilised strength was between 0.2τpeak and 0.8τpeak. Exponents of the power–law functions of these sands were found to be lower than in the published undrained shearing data of clays. Using γM=2 as the reference strain shows a slightly better power–law correlation than using γpeak. Linear relationships between the reference strains and variables, such as relative density, relative dilatancy index, and dilatancy, are identified. Full article

Aiming to resolve the problem of the poor peanut seed-filling effect under high-speed operation when developing high-speed peanut sowing with precision, a peanut precision seed-metering machine with an auxiliary air-suction seed-filling device was designed. Focusing on the force analysis of peanuts in the seed chamber, the peanut seed disturbance principle in the seed-metering machine for the blowing structure of an auxiliary air-suction seed-filling device was clarified. The seed-filling process was analyzed via DEM-CFD coupled simulation, and three factors affecting the seed-filling effect were identified, namely the seed-filling chamber ‘V’ angle γ, the bottom blow-air-hole cross-sectional area S, and the bottom blow-air-hole airflow velocity vq, and the ranges of values of the three factors were determined. The Box–Behnken test was conducted using the seed-filling index and leakage index as the indexes. The results show that the seed-filling chamber ‘V’ angle γ is 56.59°, the bottom blowhole cross-sectional area S is 1088.4 mm2, and the blowhole air velocity vq is 12.11 m·s−1. At this point, the peanut seed suction qualification index and leakage index are optimal, the seed suction qualification index is 96.33%, and the seed leakage index is 2.59%. At the same time, the field test shows that a sowing operation speed of 8–12 km·h−1, a qualified index > 93%, and a leakage index < 4.5% are required to meet the agronomic requirements of peanut precision sowing. PSR B1259-63/LS 2883 is a well-studied gamma-ray binary hosting a pulsar in a 3.4-year eccentric orbit around a Be-type star. Its non-thermal emission spans from radio to TeV energies, exhibiting a significant increase near the periastron passage. This paper is dedicated to the analysis of INTEGRAL observations of the system following its last periastron passage in June 2024. We aim to study the spectral evolution of this gamma-ray binary in the soft (0.3–10 keV) and hard (30–300 keV) X-ray energy bands. We performed a joint analysis of the data taken by INTEGRAL/ISGRI in July–August 2024 and quasi-simultaneous Swift/XRT observations. The spectrum of the system in the 0.3–300 keV band is well described by an absorbed power law with a photon index of Γ=1.42±0.03. We place constraints on potential spectral curvature, limiting the break energy Eb>30 keV for ΔΓ>0.3 and cutoff energy Ecutoff>150 keV at a 95% confidence level. For one-zone leptonic emission models, these values correspond to electron distribution spectral parameters of Eb,e>0.8 TeV and Ecutoff,e>1.7 TeV, consistent with previous constraints derived by H.E.S.S. The development of scalable, non-invasive tools to assess tumor responsiveness to structurally active immunoformulations remains a critical unmet need in solid tumor immunotherapy. Here, we introduce a real-time, ex vivo functional system to classify tumor cell lines exposed to a phospholipoproteomic platform, without relying on cytotoxicity, co-culture systems, or molecular profiling. Tumor cells were monitored using IncuCyte® S3 (Sartorius) real-time imaging under ex vivo neutral conditions. No dendritic cell components or immune co-cultures were used in this mode. All results are derived from direct tumor cell responses to structurally active formulations. Using eight human tumor lines, we captured proliferative behavior, cell death rates, and secretomic profiles to assign each case into stimulatory, inhibitory, or neutral categories. A structured decision-tree logic supported the classification, and a Functional Stratification Index (FSI) was computed to quantify the response magnitude. Inhibitory lines showed early divergence and high IFN-γ/IL-10 ratios; stimulatory ones exhibited a proliferative gain under balanced immune signaling. The results were reproducible across independent batches. This system enables quantitative phenotypic screening under standardized, marker-free conditions and offers an adaptable platform for functional evaluation in immuno-oncology pipelines where traditional cytotoxic endpoints are insufficient. This approach has been codified into the STIP (Structured Traceability and Immunophenotypic Platform), supporting reproducible documentation across tumor models. This platform contributes to upstream validation logic in immuno-oncology workflows and supports early-stage regulatory documentation. Full article

In unmanned systems, especially in large-scale and complex ones, sensor and communication failures occur from time to time and are hard to avoid. Therefore, this paper studies the fault detection problem of a class of unknown nonlinear singular uncertain time-varying delay Markov jump systems (UNSUTVDMJSs). Firstly, the corresponding sliding mode controller (SMC) is designed by using the equivalent control principle, and the unknown nonlinearity is equivalently replaced by changing the system input. Then, a fault detection filter adapted to this system is designed, thereby obtaining the unknown nonlinear stochastic singular uncertain Augmented filter residual system (UNSSUAFRS) model. To obtain the sufficient conditions for the random admissibility of this augmented system, a weak infinitesimal generator was used to design the required Lyapunov-Krasovskii functional. With the help of the Lyapunov principle and H performance analysis method, the sufficient conditions for the random admissibility of UNSSUAFRS under the H performance index γ were derived. Finally, with the aid of the designed residual evaluation function and threshold, simulation analysis was conducted on the examples of DC servo motors and numerical calculation examples to verify the effectiveness and practicability of this fault detection filter. Investigating the construction of greenway network systems in mountainous suburban areas from an integrated “ecology–recreation” perspective is crucial for promoting the coordinated development of regional multifunctionality. Taking Jinma Mountain in Kunming as a specific case study, this research comprehensively adopts a multivalue, multidimensional perception evaluation method to construct an assessment framework for suburban mountainous greenway networks that couples ecological and recreational functions. The results show that the Jinma Mountain greenway network exhibits a unique “multiple rings intertwined and dense network” pattern, with an optimized density of 0.79 km/km2, achieving efficient utilization. Compared to single-function greenways, the network’s ring index (α), connectivity index (β), and cohesion index (γ) have improved by 12.88%, 20%, and 4.19%, respectively, demonstrating a high degree of coupling and coordination. These improvements demonstrate the rationality and scientific rigor of the designed evaluation system, offering significant advantages over traditional single-function greenways. This comprehensive evaluation system not only supplements existing research on greenway networks but also provides a theoretical reference for integrated “ecology–recreation” and sustainable development in mountainous suburban areas. The structures of hexaammine solvated indium(III) and thallium(III) ions in liquid ammonia solution are determined by EXAFS. Both complexes have regular octahedral coordination geometry with mean In-N and Tl-N bond distances of 2.23(1) and 2.29(2) Å, respectively. Ammine solvated thallium(III) in liquid ammonia is characterized with 205Tl NMR measurements. Solvents such as liquid ammonia, N,N-dimethylthioformamide (DMTF), trialkyl and triphenyl phosphite and phosphine are strong electron pair donors and thereby able to form bonds with a large covalent contribution with strong electron pair acceptors. A survey of reported structures of ammine, DMTF, trialkyl and triphenyl phosphite and phosphine solvated metal ions in the solid state and solution is presented. The M-N and M-S bond distances in ammine and DMTF solvated metal ions are compared with the M-O bond distance in the corresponding metal ion hydrates, expected to form mainly electrostatic interactions with metal ions. The d10 metal ions have high ability to form bonds with a high degree of covalency with increasing ability down the group and with decreasing charge of the metal ion. The difference in M-N and M-O bond distances between ammine solvated and hydrated metal ions with the same coordination geometry decreases significantly with the increasing ability of the metal ion to form bonds with a large covalent contribution. This difference correlates well with the covalent bonding index, γM2*r. Full article

Fusaric acid (FSA) is a mycotoxin produced by pathogenic Fusarium species that inhibits the growth of various beneficial microbes. In this study, we investigated the molecular mechanisms by which Trichoderma harzianum NJAU4742 (Th), a beneficial fungus, responds to FSA-induced stress. Here, by combining a transcriptome analysis, a gene knockout, and physiological data measurements, our study investigated the molecular mechanisms underlying the response of Trichoderma harzianum NJAU4742 (Th) to FSA stress. The results showed that FSA can induce severe oxidative stress in Th, and an aldehyde dehydrogenase (Thaldh3) in Th plays a critical role in alleviating FSA stress. Deleting Thaldh3 significantly decreased the γ-aminobutyrate (GABA) content, causing more severe oxidative damage in Th. Furthermore, we also provide evidence demonstrating that Thaldh3 alleviates FSA stress by enhancing the activities of key enzymes involved in the tricarboxylic acid cycle and ATP content. A pot experiment showed that an enhanced tolerance to FSA increased the Th biomass, strengthening its antagonistic capacity against pathogens and reducing the disease index in tomatoes. In conclusion, these observations provide new insight into the role of beneficial microbes in promoting plant health. Full article

Estrogen-related receptor γ (ERRγ) has been reported to regulate various inflammation-related diseases. Herein, we attempted to evaluate the effects of DN200434 as a modulator for ERRγ in mice with atopic dermatitis (AD). Levels of mRNA and protein expression for ERRγ were evaluated in normal and DNCB-induced AD-diagnosed skin. The effects of DN200434 on the chemokines, inflammatory cytokines, and AKT/MAPK/NFκB pathway signaling were investigated in TNF-α/IFN-γ-treated HaCaT cells. DNCB-induced AD mice received DN200434 intraperitoneally for 10 days. Epidermal thickness at the dorsal aspect of the inflamed skin, spleen index, serum IgE levels, and proinflammatory cytokine levels in the skin lesions were measured. Histopathological evaluations, including assessments of epidermal hyperplasia, dermal inflammation, hyperkeratosis, folliculitis, and mast cell counts, were performed to confirm diagnostic features. Significant elevations in ERRγ expression at the RNA and protein levels were observed in DNCB-induced AD lesions. DN200434 suppressed chemokine and inflammatory cytokine expression and inhibited the elevated phosphorylation levels of AKT, ERK, p38, and NFκB in TNF-α/IFN-γ-treated HaCaT cells. Treatment with DN200434 alleviated DNCB-induced AD symptoms. The histopathological score and levels of infiltrated mast cells were also markedly lower in DN200434-treated AD mice than in vehicle-treated AD mice. Consistently, DN200434 reduced the serum IgE level and mRNA expression of TNFα and IL-6 in AD-diagnosed lesions. Collectively, our findings indicated the feasibility of ERRγ as a therapeutic target for the regulation of AD and that DN200434 can be a useful therapeutic agent in treating AD. Full article

Free space optical communication (FSOC) systems operating in the space–atmosphere channel are susceptible to severe turbulence-induced scintillation, particularly in up-link configurations where the adaptive optics (AO) pre-correction becomes ineffective due to anisoplanatic constraints. This study presents a novel scintillation suppression strategy utilizing self-focusing optical pin-like beams (OPBs) with tailored phase modulation, combining theoretical derivation and numerical simulation. It is found that increasing the shape factor γ and modulation depth C elevates the average received power and reduces the scintillation index at the focal point. Meanwhile, quantitative evaluation of the five OPB configurations shows that the parameter set γ = 1.4 and C = 7 × 10−5 gives a peak scintillation suppression efficiency. It shows that turbulence induced scintillation is suppressed by 44% with the turbulence intensity D/r0 = 10, demonstrating exceptional effectiveness in up-link transmission. The findings demonstrate that OPB with optimized γ and C establish an approach for uplink FSOC, which is achieved through suppressed scintillation and stabilized power reception. Full article

Previously, research adopted an ultrasound-assisted extraction method to isolate crude polysaccharide from blackened jujube, followed by preliminary structural identification of the purified polysaccharide (BJP). This manuscript analyzed the accurate structure and immunomodulatory activity of BJP. Further structural identification indicated that BJP was mainly composed of →3)-α-L-Araf-(1→, →3,5)-α-L-Araf-(1→, →3)-β-D-GalpA-(1→, →2,4)-β-D-Galp-(1→, →4)-β-D-GalpA-(1→, →3)-α-L-Rhap-(1→ and →3,4)-α-L-Rhap-(1→. The immunomodulatory effects of BJP were examined using a mouse model with immunosuppression induced by cyclophosphamide. The findings suggested that BJP could relieve the condition of immunosuppressed mice. BJP could inhibit decreases in the body weight and organ index of mice, and HE staining showed that BJP could alleviate the harm to spleen and thymus tissues. BJP enhanced the secretion of interferon-γ (IFN-γ), tumor necrosis factor-α (TNF-α), interleukin-2 (IL-2), interleukin-6 (IL-6), immunoglobulin A (IgA), and immunoglobulin G (IgG) in serum. It also reduced liver oxidative stress by increasing superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) activities, while lowering malondialdehyde (MDA) levels. Moreover, BJP contributed to the maintenance of gut homeostasis by stimulating the generation of short-chain fatty acids in the cecal contents. The study aims to establish a solid basis for the comprehensive development of blackened jujube and furnish a theoretical framework for its polysaccharides’ role in immune modulation. Full article

The therapeutic target of COVID-19 is focused on controlling inflammation and preventing fibrosis. Collagen–polyvinylpyrrolidone (collagen-PVP) and pirfenidone both have the ability to control the cytokine storm observed in rheumatic and fibrotic disorders. In this work, our aim was to understand the benefits of treatment with each of these drugs in patients with severe COVID-19. In total, 36 patients were treated with dexamethasone and enoxaparin, but 26 were allocated collagen-PVP or pirfenidone (n = 15 and 11, respectively); the clinical and metabolic effects were compared among them. Since pirfenidone works via transcriptional mechanisms, we performed a human genome microarray assay using RNA isolated from fibroblast and monocyte cultures treated with the biodrug, with the aim of hypothesising a possible mechanism of action for collagen-PVP. Our results showed that hospital stay duration, quick COVID-19 severity index (qCSI), and admission to the intensive care unit were statistically significantly lower (p < 0.02) in patients treated with collagen-PVP or pirfenidone when compared with the control group, and that only collagen-PVP normalised serum glucose at discharge. Ingenuity Pathway Analysis showed that the cell cycle, inflammation, and cell surface–extracellular matrix interactions could be regulated with collagen-PVP via the downmodulation of proinflammatory cytokines, while Th2 anti-inflammatory response signalling could be upregulated. Furthermore, the downregulation of some of the genes involved in nitric oxide production showed a possible control for JAK in the IFN-γ pathway, allowing for the possibility of controlling inflammation through the JAK/STAT pathway, as has been observed for pirfenidone and other immunomodulators, such as ruxolitinib. Background/Objectives: Myopia is associated with peripapillary changes, namely, gamma peripapillary atrophy (γPPA) and optic disc ovalization, estimated by the ovality index (OI). These changes have been suggested to be promoted by adduction. Recent studies highlight that near reading significantly contributes to the development and progression of myopia and that the interpupillary distance (IPD) influences vergence amplitudes. While both adduction and convergence are involved during near reading, a potential link between IPD and myopic peripapillary changes has not yet been explored. We, therefore, sought to determine whether IPD is related to the OI or γPPA width. Methods: In this monocentric cross-sectional study, 100 eyes from 100 adults (mean age of 62.6 ± 13.7 years) were analyzed. Axial length (AL), refractive error, and IPD were recorded. The OI and γPPA width were assessed using spectral-domain Optical Coherence Tomography. Pearson correlations and multivariable linear regressions were performed, adjusting for age, gender, and myopia status. Results: IPD showed no significant correlation with the OI (r = 0.001; p = 0.989) or γPPA (r = −0.028; p = 0.789). A weak, non-significant correlation was found between IPD and AL (p = 0.059). In contrast, AL was strongly correlated with both a lower OI and wider γPPA (p < 0.001). Conclusions: These findings suggest that IPD-related biomechanical forces do not influence optic nerve head (ONH) shape or γPPA. Axial elongation remains the key driver of myopic ONH remodeling. Background/Objectives: Lactobacillus johnsonii CRL1231 (Lj CRL1231) is a strain with feruloyl esterase (FE) activity that enhances ferulic acid (FA) release from wheat bran (WB) and has potential as a probiotic for metabolic syndrome (MS). Given the potential health benefits of FA and its microbial metabolites, this study aimed to evaluate the therapeutic effect of Lj CRL1231 co-administered with WB in a mouse model of metabolic syndrome (MS) induced by a high-fat diet (HFD). Methods: Mice were divided into three groups and fed for 14 weeks as follows: the Control group (standard diet), the MS group (HFD+WB), and the MS+Lj group (HFD+WB and Lj CRL1231-dose 108 cells/day). Specifically, we analyzed the changes in the intestinal microbiota (IM), colonic FE activity, generation of FA-derived and fermentation metabolites, and metabolic and inflammatory parameters. Results: Improvements in the MS+Lj group compared to the MS group included the following: a—a 38% increase in colonic FE activity, leading to elevated levels of FA-derived metabolites (e.g., dihydroferulic, dihydroxyphenylpropionic, and hydroxyphenylpropionic acids); b—a significant shift in the IM composition, with a 3.4-fold decrease in Firmicutes and a 2.9-fold increase in Bacteroidetes; c—a decrease in harmful bacteria (Desulfovibrio) by 93%, and beneficial bacteria like Bifidobacterium increased significantly (6.58 log cells/g); d—a 33% increase in total SCFAs; e—a 26% reduction in the adiposity index; f—a 12% increase in HDL cholesterol and a 19% reduction in triglycerides; g—normalized glucose and insulin resulting in a 2-fold lower HOMA-IR index; h—an improved inflammatory profile by decreasing TNF-α, IFN-γ, and IL-6 (3-, 5-, and 2-fold, respectively) and increasing IL-10 by 2-fold; i—alleviation of liver damage by normalizing of transaminases AST (19.70 ± 2.97 U/L) and ALT (13.12 ± 0.88 U/L); j—evidence of reduced oxidative damage. Conclusions: The co-administration of L. johnsonii CRL1231 and WB exerts a synergistic effect in mitigating the features of MS in HFD-fed mice. This effect is mediated by modulation of the gut microbiota, increased release of bioactive FA-derived compounds, and restoration of metabolic and inflammatory homeostasis. This strategy represents a promising dietary approach for MS management through targeted microbiota–metabolite interactions. Full article

Vitamin D deficiency is highly prevalent in individuals with overweight/obesity and this can be largely attributed to the entrapment of VitD in adipose tissue due to impaired lipolytic stimulation. Considering the well-described role of exercise in stimulating lipolysis, the present study investigated the efficacy of multicomponent-type high-intensity interval training (m-HIIT) in increasing 25-hydroxyvitamin D [25(OH)D] levels in males with overweight/obesity. Twenty middle-aged males (43.5 ± 5 years, BMI: 30.7 ± 3.3 kg/m2) participated in three weekly supervised m-HIIT sessions over a 12-week period and underwent assessments at baseline, 6, and 12 weeks. Primary outcomes were total body fat mass, android fat, hepatorenal index, and serum 25(OH)D. Participants’ daily physical activity and dietary intake habits remained unaltered throughout the 12-week training period. The m-HIIT intervention reduced fat mass (by 3% at 12 weeks), android fat (by 3.7% at 6 weeks and 4.4% at 12 weeks), and hepatorenal index (by 8% at 12 weeks). Serum 25(OH)D levels increased by ~14% (+3.21 ng/mL, p = 0.002) and ~31% (+7.24 ng/mL, p < 0.001) at 6 and 12 weeks, respectively. The elevation of 25(OH)D levels at 12 weeks was inversely related to fat mass loss (R = 0.53, p = 0.016). Plasma SGPT, SGOT, ALP, γ-GT, fetuin-A, and calcium levels remained unaltered after the 12-week training period. In conclusion, m-HIIT may be useful as a non-pharmacological intervention to increase circulating VitD levels in adults with overweight/obesity. Full article
