Dear Colleagues,
Heart failure (HF) is a complex clinical syndrome that affects an increasing proportion of the population; as such, it remains a major therapeutic challenge. Despite significant progress in pharmacological and device-based therapies, morbidity and mortality rates remain substantial and are closely related to the severity of the disease. Overall, the five-year mortality rate is approximately 50%, which is comparable to many malignancies.
For this reason, the search for new therapeutic strategies in the treatment of HF has become an urgent priority in the current clinical landscape.
Understanding molecular mechanisms and cellular signaling pathways is essential both for elucidating the physiology of the cardiovascular system and for identifying novel therapeutic strategies in HF. At the physiological level, these pathways regulate fundamental processes such as myocardial contractility, energy metabolism, calcium homeostasis, etc. Disruptions in these finely tuned mechanisms contribute directly to the development and progression of heart failure.
From a therapeutic perspective, detailed knowledge of molecular signaling enables the identification of key regulatory nodes that can be selectively targeted by pharmacological or genetic interventions. This not only facilitates the development of more precise and effective treatments but also allows for a shift from symptomatic management toward mechanism-based therapy. Consequently, advances in the understanding of molecular and signaling pathways are critical for improving outcomes and enabling the development of innovative, targeted approaches in the treatment of HF.
