Cover Story: Understanding the mechanisms governing prolonged drug retention at target sites is vital for enhancing treatment efficacy. In this Special Issue, Sharma et al. investigate the dynamic interaction of epichaperome agents, such as zelavespib and icapamespib, with the target in vivo, unveiling the intricate processes that contribute to their extended retention at the target site and their effectiveness in treating various diseases. The cover highlights the pivotal role of specific binding and stabilization within the epichaperome complex, leading to target disruption.
