In the first place, the relationship between  SNPs and MRONJ is not major. Therefore, it is insufficient explanation to this relation in Introduction or Discussion. The authors should add more described why did you write this review.

The authors described "This study mainly highlights ~~~reduce the possibility of developing MRONJ. in Conclusions.  What do the authors think about How to reduce the possibility of developing MRONJ?  Because oral surgeons have to drive down occurrence frequency of MRONJ rate.

Answer: Thanks for your remark. This has now been added in the Discussion section. We have also highlighted the meaning on this review by adding a paragraph at the end of the Introduction.

I am so delighted that read and review your valuable article. I have some suggestions which would help for more clarification.

Abstract:

1. In the abstract, the goal of your study is decreased to " summarizing the results of ...", but as you stated at the end of introduction, you appraised the results of previous secondary studies. I think the abstract can be edited.

 

Introduction:

2. In my opinion, the table 1 is unnecessary, because the goal of your study is about the role of pharmacogenomic aspects of MRONJ  and this table has caused the introduction to be long. The reader is eager to know more about the genetic and pharmacogenomic aspect of MRONJ.  

 

Materials and Methods:

-It is recommended to cite the last edition of PRISMA (2020).

-Please use RISMA 2020 flow diagram template.

-Why did not place "BRONJ" in the key words for searching the data base?

-Please add the restriction regarding the year of publication.

-For data extraction,Was the concordance between 5 reviewers assessed?

Answer: Thanks for your remark. The abstract has been edited. Two sentences have been added and one has been deleted.

Answer: Thanks for your remark. The PRISMA reference and diagram template have been changed. We ran a new search by adding BRONJ as key words which did not add any new reviews. The restriction regarding year of publication has now been added. The data extraction was agreed among all the 5 assessor reviewers; in case of disagreement a discussion was conducted until consensus was obtained.

This review entitled 'A comprehensive quality meta-review of genetic and pharmacogenomic aspects of medication-related osteonecrosis of the jaw (MRONJ)' appears to deal with a very interesting subject about which there is still little knowledge and certainty, as is also evident from the conclusions of the work under review. MRONJ is a condition that has become a serious medical problem over the last few decades, partly as a result of the ever-increasing spread of drugs that can induce it, which is why a review that seeks to take stock of the current state of knowledge on how to prevent its onset is very useful for both researchers and clinicians. 

I congratulate the authors as the work is well written and structured, delving into the many aspects that could come into play in the development of MRONJ, however I feel that some minor improvements could further enhance the quality of the work. 

Introduction: 

- It is too long and risks forcing readers, focus on the concepts strictly necessary for the introduction of the work

- The introduction should not contain tables. Please delete table number 1, also because it does not provide any useful information for understanding the work. 

- Please better specify the objectives of the study, emphasising the authors' reasons for wanting to write the following review 

Discussion 

- It is too long. Focus on the concepts essential to demonstrate how there is currently no unanimous consensus on the predisposition that the expression of certain genes might have on the development of MRONJ. 

Conclusions

Add a short paragraph explaining what the authors think future research should look into in order to increase current knowledge on the topic.

Answer: Thanks for your remark. The discussion has been shortened by removing two paragraphs. However, we had to add a new paragraph as per another reviewer’s comment. We also believe that the Discussion contains information related to the contradictions of the many studies included in the reviews appraised. In fact, none of the genome-wide association studies (GWAS), candidate gene studies (CGS), whole genome and whole exome studies have reported a strong correlation between genetic and MRONJ.

Conclusions

Add a short paragraph explaining what the authors think future research should look into in order to increase current knowledge on the topic.
