Cover Story: In this work, the specificity of human immunodeficiency virus type 1 (HIV-1) protease was studied in vitro and in silico, in the context of the cleavage site representing the sequence of the proximal zinc finger of HIV-1 nucleocapsid. The results revealed a preference for large and hydrophobic residues in the P1′ position and the importance of alternative binding possibilities of substrates. This paper is dedicated to the loving memory of Stephen Oroszlan, who made an essential contribution to the field of retrovirology; the presented results are based on his ideas and follow his inspirations.
