Optimization of the Viability of Microencapsulated Lactobacillus reuteri in Gellan Gum-Based Composites Using a Box–Behnken Design

The growing interest in probiotic bacteria within the food industry is driven by their recognized health benefits for consumers. However, preserving their therapeutic viability and stability during gastrointestinal transit remains a formidable challenge. Hence, this research aimed to enhance the viability of Lactobacillus reuteri through microencapsulation using a binary polysaccharide mixture composed of low acyl gellan gum (LAG), high acyl gellan gum (HAG), and calcium for the microencapsulation of L. reuteri. To achieve this, the Box–Behnken design was applied, targeting the optimization of L. reuteri microencapsulated to withstand simulated gastrointestinal conditions. The microcapsules were crafted using the internal ionic gelation method, and optimization was performed using response surface methodology (RSM) based on the Box–Behnken design. The model demonstrated robust predictive power, with R2 values exceeding 95% and a lack of fit greater than p > 0.05. Under optimized conditions—0.88% (w/v) LAG, 0.43% (w/v) HAG, and 24.44 mM Ca—L. reuteri reached a viability of 97.43% following the encapsulation process. After 4 h of exposure to simulated gastric fluid (SGF) and intestinal fluid (SIF), the encapsulated cells maintained a viable count of 8.02 log CFU/mL. These promising results underscore the potential of biopolymer-based microcapsules, such as those containing LAG and HAG, as an innovative approach for safeguarding probiotics during gastrointestinal passage, paving the way for new probiotic-enriched food products. This review explores the biofilm architecture and drug resistance of Enterococcus faecalis in clinical and environmental settings. The biofilm in E. faecalis is a heterogeneous, three-dimensional, mushroom-like or multilayered structure, characteristically forming diplococci or short chains interspersed with water channels for nutrient exchange and waste removal. Exopolysaccharides, proteins, lipids, and extracellular DNA create a protective matrix. Persister cells within the biofilm contribute to antibiotic resistance and survival. The heterogeneous architecture of the E. faecalis biofilm contains both dense clusters and loosely packed regions that vary in thickness, ranging from 10 to 100 µm, depending on the environmental conditions. The pathogenicity of the E. faecalis biofilm is mediated through complex interactions between genes and virulence factors such as DNA release, cytolysin, pili, secreted antigen A, and microbial surface components that recognize adhesive matrix molecules, often involving a key protein called enterococcal surface protein (Esp). Clinically, it is implicated in a range of nosocomial infections, including urinary tract infections, endocarditis, and surgical wound infections. The biofilm serves as a nidus for bacterial dissemination and as a reservoir for antimicrobial resistance. The effectiveness of first-line antibiotics (ampicillin, vancomycin, and aminoglycosides) is diminished due to reduced penetration, altered metabolism, increased tolerance, and intrinsic and acquired resistance. Alternative strategies for biofilm disruption, such as combination therapy (ampicillin with aminoglycosides), as well as newer approaches, including antimicrobial peptides, quorum-sensing inhibitors, and biofilm-disrupting agents (DNase or dispersin B), are also being explored to improve treatment outcomes. Environmentally, E. faecalis biofilms contribute to contamination in water systems, food production facilities, and healthcare environments. They persist in harsh conditions, facilitating the spread of multidrug-resistant strains and increasing the risk of transmission to humans and animals. Therefore, understanding the biofilm architecture and drug resistance is essential for developing effective strategies to mitigate their clinical and environmental impact. Full article

Sulfur-containing fertilizers increase production costs, which leads to low utilization of this nutrient. Thus, evaluating how the absence of sulfur influences the early development of Urochloa brizantha is essential. Study was conducted in a greenhouse at the Federal University of Rondonópolis in a completely randomized design, with six treatments in a 3 × 2 factorial scheme, and eight replications. Three cultivars of U. brizantha (Marandu, Xaraés and Piatã) were evaluated under two fertilization strategies: with or without sulfur fertilization. Sufur presence increased the number of leaves and forage mass, in which cultivar Xaraés presented the greatest means. Piatã was the cultivar most sensitive to sulfur deficiency at establishment, which reduced forage mass, number of leaves and number of tillers by 42%, 32%, and 45%, respectively. Despite these differences between cultivars, sulfur efficiently increased the forage yield. Sulfur fertilization increased the concentrations of nutrients in the plants without significantly affecting the uptake of nitrogen, phosphorus, potassium, calcium and magnesium. Sulfur omission resulted in increased phosphorus uptake in all grass. In contrast, Marandu grass exhibited the greatest reduction in sulfur uptake. Therefore, the use of sulfur in the fertilization of grasses is recommended, it is important to evaluate the responses of each cultivar to better adjust the fertilization management. Full article

This study aims to evaluate eco-innovative solutions in the fertilizer industry that allow for waste valorization in the context of a resource-efficient circular economy. A comprehensive reuse strategy was developed for low-rank peat and post-cultivation horticultural mineral wool, involving the extraction of valuable humic substances from peat and residual nutrients from used mineral wool, followed by the use of both post-extraction residues to produce organic–mineral substrates. The resulting products/semifinished products were characterized in terms of their composition and properties, which met the requirements necessary to obtain the admission of this type of product to the market in accordance with the Regulation of the Minister for Agriculture and Rural Development of 18 June 2008 on the implementation of certain provisions of the Act on fertilizers and fertilization (Journal of Laws No 119, item 765). Elemental analysis, FTIR spectroscopy, and solid-state CP-MAS 13C NMR spectroscopy suggest that post-extraction peat has a relatively condensed structure with a high C content (47.4%) and a reduced O/C atomic ratio and is rich in alkyl-like matter (63.2%) but devoid of some functional groups in favor of extracted fulvic acids. Therefore, it remains a valuable organic biowaste, which, in combination with post-extraction waste mineral wool in a ratio of 60:40 and possibly the addition of mineral nutrients, allows us to obtain a completely new substrate with a bulk density of 264 g/m3, a salinity of 7.8 g/dm3 and a pH of 5.3, with an appropriate content of heavy metals and with no impurities, meeting the requirements of this type of product. A liquid fertilizer based on an extract containing previously recovered nutrients also meets the criteria in terms of quality and content of impurities and can potentially be used as a fertilizing product suitable for agricultural crops. This study demonstrates a feasible pathway for transforming specific waste streams into valuable agricultural inputs, contributing to environmental protection and sustainable production. The production of a new liquid fertilizer using nutrients recovered from post-cultivation mineral wool and the preparation of an organic–mineral substrate using post-extraction solid residue is a rational strategy for recycling hard-to-biodegrade end-of-life products. Bamboo shoot boiled liquid (BSBL), a processing byproduct containing soluble proteins, peptides, amino acids, carbohydrates, and phenolics, is typically discarded, causing resource waste and environmental issues. This study analyzed metabolic changes in BSBL during Pediococcus pentosaceus B49 fermentation. The result of partial least squares discriminant analysis (PLS-DA) revealed significant metabolite profile differences across fermentation times (0 h, 24 h, 48 h, 72 h, 96 h). The most substantial alterations occurred within the first 24 h, followed by stabilization. Compared to unfermented BSBL, fermented samples exhibited significantly elevated signal intensities for 5,7-dimethoxyflavone, cinnamic acid, 3,4-dihydro-2H-1-benzopyran-2-one, 6,8-dimethyl-4-hydroxycoumarin, and 2-hydroxycinnamic acid (p < 0.05), showing upward trends over time. Conversely, (+)-gallocatechin intensity decreased gradually. Bitter peptides, such as alanylisoleucine, isoleucylisoleucine, leucylvaline, and phenylalanylisoleucine, in BSBL exhibited a significant reduction following fermentation with P. pentosaceus B49 (p < 0.05). KEGG enrichment indicated tyrosine metabolism (ko00350) and arginine/proline metabolism (ko00330) as the most impacted pathways. These findings elucidate metabolic regulation in BSBL fermentation, supporting development of functional fermented bamboo products. Full article

Burdock (Arctium lappa L.) is a medicinal and edible homologous plant whose roots contain many bioactive substances such as polysaccharides and phenolics. This study explored the integration of burdock powder and lactic acid bacteria fermentation to enhance the nutritional quality, sensory attributes, and flavor profiles of duck sausages. Three bacterial strains, Lacticaseibacillus casei, L. helveticus, and L. plantarum, were selected based on sensory analysis, and their effects on sausage properties were evaluated through combined fermentation trials. The results demonstrated that duck sausages fermented with L. plantarum and L. helveticus and supplemented with 3% burdock powder (PHB group) exhibited > 1.5-fold higher antioxidant activity (ABTS at 85.2 μmol trolox/g and DPPH at 92.7 μmol trolox/g, respectively; p < 0.05) and 15% increase in total phenolic content (8.24 mg gallic acid/g) compared to non-fermented counterparts. The PHB formulation also enhanced color stability (lightness, redness, yellowness), textural characteristics (hardness, springiness, cohesiveness), and sensory acceptability. Volatile compound analysis revealed a reduction in off-odor aldehydes (hexanal, (E)-2-octenal, (E)-2-decenal, and (E,E)-2,4-decadienal) and increased production of desirable aromatic compounds like tetramethyl-pyrazine. These findings highlight the potential of combining lactic acid bacteria fermentation with burdock powder to develop functional duck sausages with improved nutritional and sensory properties. Contact unmodified antisense DNA biotechnology (CUADb), developed in 2008, employs short antisense DNA oligonucleotides (oligos) as a novel approach to insect pest control. These oligonucleotide-based insecticides target pest mature rRNAs and/or pre-rRNAs and have demonstrated high insecticidal efficacy, particularly against sap-feeding insect pests, which are key vectors of plant DNA viruses and among the most economically damaging herbivorous insects. To further explore the potential of CUADb, this study evaluated the insecticidal efficacy of short 11-mer antisense DNA oligos against Coccus hesperidum, in comparison with long 56-mer single-stranded and double-stranded DNA sequences. The short oligos exhibited higher insecticidal activity. By day 9, the highest mortality rate (97.66 ± 4.04%) was recorded in the Coccus-11 group, while the most effective long sequence was the double-stranded DNA in the dsCoccus-56 group (77.09 ± 6.24%). This study also describes the architecture of the DNA containment (DNAc) mechanism, highlighting the intricate interactions between rRNAs and various types of DNA oligos. During DNAc, the Coccus-11 treatment induced enhanced ribosome biogenesis and ATP production through a metabolic shift from carbohydrates to lipid-based energy synthesis. However, this ultimately led to a ‘kinase disaster’ due to widespread kinase downregulation resulting from insufficient ATP levels. All DNA oligos with high or moderate complementarity to target rRNA initiated hypercompensation, but subsequent substantial rRNA degradation and insect mortality occurred only when the oligo sequence perfectly matched the rRNA. Both short and long oligonucleotide insecticide treatments led to a 3.75–4.25-fold decrease in rRNA levels following hypercompensation, which was likely mediated by a DNA-guided rRNase, such as RNase H1, while crucial enzymes of RNAi (DICER1, Argonaute 2, and DROSHA) were downregulated, indicating fundamental difference in molecular mechanisms of DNAc and RNAi. Consistently, significant upregulation of RNase H1 was detected in the Coccus-11 treatment group. In contrast, treatment with random DNA oligos resulted in only a 2–3-fold rRNA decrease, consistent with the normal rRNA half-life maintained by general ribonucleases. These findings reveal a fundamental new mechanism of rRNA regulation via complementary binding between exogenous unmodified antisense DNA and cellular rRNA. From a practical perspective, this minimalist approach, applying short antisense DNA dissolved in water, offers an effective, eco-friendly and innovative solution for managing sternorrhynchans and other insect pests. The results introduce a promising new concept in crop protection: DNA-programmable insect pest control. Full article

Optimizing Multi-Cluster Fracture Propagation and Mitigating Interference Through Advanced Non-Uniform Perforation Design in Shale Gas Horizontal Wells

The persistent challenge of fracture-driven interference (FDI) during large-scale hydraulic fracturing in the southern Sichuan Basin has severely compromised shale gas productivity, while the existing research has inadequately addressed both FDI risk reductions and the optimization of reservoir stimulation. To bridge this gap, this study developed a mechanistic model of the competitive multi-cluster fracture propagation under non-uniform perforation conditions and established a perforation-based design methodology for the mitigation of horizontal well interference. The results demonstrate that spindle-shaped perforations enhance the uniformity of fracture propagation by 20.3% and 35.1% compared to that under uniform and trapezoidal perforations, respectively, with the perforation quantity (48) and diameter (10 mm) identified as the dominant control parameters for balancing multi-cluster growth. Through a systematic evaluation of the fracture communication mechanisms, three distinct inter-well types of FDI were identified: Type I (natural fracture–stress anisotropy synergy), Type II (natural-fracture-dominated), and Type III (stress-anisotropy-dominated). To mitigate these, customized perforation schemes coupled with geometry-optimized fracture layouts were developed. The surveillance data for the offset well show that the pressure interference decreased from 14.95 MPa and 6.23 MPa before its application to 0.7 MPa and 0 MPa, achieving an approximately 95.3% reduction in the pressure interference in the application wells. The expansion morphology of the inter-well fractures confirmed effective fluid redistribution across clusters and containment of the overextension of planar fractures, demonstrating this methodology’s dual capability to enhance the effectiveness of stimulation while resolving FDI challenges in deep shale reservoirs, thereby advancing both productivity and operational sustainability in complex fracturing operations. The growing demand for natural and sustainable skincare products has driven interest in plant-based active ingredients, especially from in vitro cultures. This placebo-controlled study investigated the impact of a facial cream containing 2% Kickxia elatine (L.) Dumort cell suspension culture extract on various skin biophysical parameters. The cream was applied to the cheek once daily for six weeks on 40 healthy female volunteers between the ages of 40 to 49. The evaluated skin parameters including skin hydration, transepidermal water loss (TEWL), erythema intensity (EI), melanin intensity (MI), skin surface pH, and skin structure, wrinkle depth, vascular lesions, and vascular discolouration. The results indicated that significant improvements were observed in skin hydration (from 40.36 to 63.00 AU, p < 0.001) and there was a decrease in TEWL score (14.82 to 11.76 g/h/m2, p < 0.001), while the skin surface pH was maintained (14.82 to 11.76 g/h/m2, p < 0.001). Moreover, the K. elatine cell extract significantly improved skin structure values (9.23 to 8.50, p = 0.028), reduced vascular lesions (2.72 to 1.54 mm2, p = 0.011), and lowered skin discolouration (20.98% to 14.84%, p < 0.001), indicating its moisturising, protective, brightening, and soothing properties. These findings support the potential use of K. elatine cell extract in dermocosmetic formulations targeting dry, sensitive, or ageing skin. Full article

During the transition of culture-featured district planning from static conservation to innovation-driven models, existing research remains constrained by mechanistic paradigms, reducing districts to functional containers and neglecting human perceptual interactions and meaning-production mechanisms. This study explores and quantifies the generative mechanisms of spatial communicability and cultural dissemination efficacy within human-centered frameworks. Grounded in humanistic urbanism, we analyze Harbin Central Street as a case study integrating historical heritage with contemporary vitality, developing a tripartite communicability assessment framework comprising perceptual experience, infrastructure utility, and behavioral dynamics. Machine learning-based threshold analysis reveals that spatial encoding elements govern communicability through significant nonlinear mechanisms. The conclusion shows synergies between street view-quantified greenery visibility and pedestrian accessibility establish critical human-centered design thresholds. Spatial data analysis integrating physiologically sensed emotional experiences and topologically analyzed spatial morphology resolves metric fragmentation while examining spatial encoding’s impact on interaction efficacy. This research provides data-driven decision support for sustainable urban renewal and enhanced cultural dissemination, advancing heritage sustainability. Full article

Skin dullness contributes to a fatigued and aged appearance, often exceeding one’s biological age. It is a common dermatological concern influenced by aging and poor lifestyle habits, regardless of ethnicity or age. This study aimed to examine advanced glycation end products (AGEs) and their receptor (receptor for AGEs [RAGE]) as contributing factors to skin dullness. AGEs themselves have a yellowish hue, contributing to “yellow dullness.” Additionally, AGE–RAGE signaling promotes melanin production in melanocytes and impairs keratinocyte differentiation as a result of inflammation. Therefore, regulating the AGE–RAGE interaction may help reduce skin dullness. Through screening various natural ingredients, we found that peony root extract (PRE) inhibits AGE formation and blocks AGE–RAGE binding. Furthermore, the presence of PRE leads to the suppression of AGE-induced melanin production in melanocytes and the restoration of impaired keratinocyte differentiation in glycated basement membrane components. In a human clinical study, topical application of a 1% PRE-containing lotion for 2 weeks significantly reduced melanin content, with a trend toward decreased AGE accumulation and visible spots on the cheeks. These findings support the potential of PRE as a multifunctional cosmetic ingredient that comprehensively addresses skin dullness by modulating the AGE–RAGE interaction. Full article

This paper investigates the cultural, spiritual, and ecological use and value of fish by-products in the material practices of Alaska Native (Indigenous Peoples are the descendants of the populations who inhabited a geographical region at the time of colonisation and who retain some or all of their own social, economic, cultural, and political institutions. In this paper, I use the terms “Indigenous” and “Native” interchangeably. In some countries, one of these terms may be favoured over the other.) and Greenlandic Inuit women. It aims to uncover how fish remnants—skins, bones, bladders, vertebrae, and otoliths—were transformed through tanning, dyeing, and sewing into garments, containers, tools, oils, glues, and adornments, reflecting sustainable systems of knowledge production rooted in Arctic Indigenous lifeways. Drawing on interdisciplinary methods combining Indigenist research, ethnographic records, and sustainability studies, the research contextualises these practices within broader environmental, spiritual, and social frameworks. The findings demonstrate that fish-based technologies were not merely utilitarian but also carried symbolic meanings, linking wearers to ancestral spirits, animal kin, and the marine environment. These traditions persisted even after European contact and the introduction of glass trade beads, reflecting continuity and cultural adaptability. The paper contributes to academic discourse on Indigenous innovation and environmental humanities by offering a culturally grounded model of zero-waste practice and reciprocal ecology. It argues that such ancestral technologies are directly relevant to contemporary sustainability debates in fashion and material design. By documenting these underexamined histories, the study provides valuable insight into Indigenous resilience and offers a critical framework for integrating Indigenous knowledge systems into current sustainability practices. Upcycling of Sunflower and Sesame Press Cakes as Functional Ingredients in Cookies

The aim of the study was to evaluate the use of sunflower and sesame oilseed press cakes, which are by-products of oil extraction, as functional ingredients in cookie production. The quality characteristics of these by-products were assessed, including water activity, pH, total phenolic content, and antioxidant activity, and HPLC analysis of the phenolic compounds was performed. Subsequently, cookies were prepared by replacing wheat flour with 30% or 50% press cake. The addition of sunflower press cake significantly increased the total phenolic content (up to 8.6 mg GAE/g dm) and antioxidant activity (up to 75.9%) in the cookies, whereas adding sesame press cake showed a less pronounced effect, reaching 0.91 g GAE/g dm and 8.9% for total phenolic content and antioxidant activity, respectively. HPLC analysis indicated that chlorogenic acid and its derivatives dominated in sunflower-enriched cookies, while sesame samples contained lignans such as sesamol and sesamin. Our study shows that 50% substitution improves the health-promoting properties of cookies and does not differ significantly from the 30% level in consumer sensory evaluations. These findings support the use of sunflower and sesame press cakes as valuable ingredients in food applications. This represents an important step toward developing healthier and more nutritious food products while supporting the principles of the circular economy through the upcycling of valuable raw materials. Phosphogypsum, a byproduct of orthophosphoric acid production, is one of the large-tonnage wastes. Since phosphogypsum mainly consists of CaSO4 2H2O, it can be considered as an alternative gypsum-bearing raw material in the production of gypsum binders. However, its features, such as particle morphology and the presence of impurities, can negatively affect the characteristics of phosphogypsum-based binders. Identification of these factors will allow us to develop methods for their minimization and increasing the efficiency of phosphogypsum use from the required source as a raw material for the production of phosphogypsum-based binders. In this regard, the manuscript contains a comprehensive and comparative analysis of phosphogypsum and natural gypsum, which makes it possible to establish their differences in chemical composition and structural and morphological features, which subsequently affect the properties of the phosphogypsum-based binder. It has been established that the key factor negatively affecting the strength of phosphogypsum-based paste (2.58 MPa) is its high water demand (0.89), which is due to the high values of the specific surface area of the particles and the presence of a large number of conglomerates with significant porosity in phosphogypsum. It has been suggested that preliminary grinding of phosphogypsum can help reduce the amount of water required to obtain fresh phosphogypsum-based paste with a standard consistency and improve its physical and mechanical properties.
